酶法合成肝靶向性脂质体载体胆固醇-半乳糖苷工艺研究

目的:利用脂肪酶催化合成肝靶向性脂质体载体胆固醇-半乳糖苷(CH-VS-LA),对其反应条件进行优化。方法:采用质谱和碳谱鉴别产物结构;单因素和正交设计法优化酶法合成条件。结果:酶法最佳条件为底物摩尔比胆固醇癸二酸乙烯酯:乳糖醇 4:1,脂肪酶Novozym435 25mg,反应32h, 产率高达 92%。结论:单因素和正交设计法优化脂肪酶Novozym 435合成脂质体载体是有效可行的。
objective: To synthesize liposomes vector, cholesterol-vinyl sebacate-lactitol (CH-VS-LA) by using enzymatic reaction and to optimize its synthesis process. Methods: MS and 13C-NMR were used to idendify the structure of product, enzymatic synthesis conditions were optimized via single-factor test and orthogonal experimental design. Results: The enzymatic optimum conditions are the followings: the molar ratio of CH-VS to LA 4:1, the amount of lipase Novozym435 25mg, reactional time 32h, productive rate 92%. Conclusion: The method that lipase Novozym435 catalyszes synthesic process of cholesterol-vinyl sebacate-lactitol by single-factor test and orthogonal experimental design is effective and feasible.
