Synthesis, characterization and controlled releasing properties of pH-responsive star polymer based on PAMAM
树形分子PAMAM通过亚胺键与疏水单元、PEG连接,制备了以PAMAM树形分子为基的pH敏感的星型聚合物PAMAM-A-PEG,通过红外光谱、核磁共振、凝胶渗透色谱/激光光散射等方法进行了表征,利用zeta电势/动态光散射联用仪对其在水溶液中的性质进行了考察,发现具有在中性条件下稳定,而在酸性条件下易于水解的pH敏感特性。以玫瑰红(RB)作为模型分子,研究了聚合物对客体分子的包覆和释放性能,结果表明PAMAM-A-PEG对于RB分子具有显著的增溶作用,这种作用来源于主客体分子间的静电作用,改变溶液pH和离子强度可以影响它们的作用强度,进而调控客体分子的释放速率。而且,PAMAM表面的疏水单元对结合在内核空腔中的客体分子的保护作用使得其具有明显的缓释效果。
pH-sensitive star polymer based on PAMAM dendrimer, PAMAM-A-PEG, was designed and synthesized via surface modification of hydrophobic unit and PEG molecules, and characterized using IR, NMR and GPC/MALLS. Its properties in solution were studied using DLS and zeta potential. It was proved that PAMAM-A-PEG had relative stability and could hydrolyze rapidly in acidic solution. Rose Bengal (RB) was used as a model guest molecule to study the encapsulating and releasing properties of the polymers. The results indicated that the star polymer could increase the resolvability of RB in some solution attributed to electrostatic interaction between host-guest molecules and the polymer, and the release behavior of RB can be regulated by adjusting the pH and ions strength in solution. In addition, the hydrophobic group on the surface of PAMAM can act as barriers to keep RB inside, and thus exhibited good retarded releasing properties.
