根据单倍体川桑基因组数据库搜索的单拷贝亚硝酸还原酶基因Morus02756设计引物,用桂优62号桑作为供试材料,以cDNA为模板,克隆桑树NiR基因的全长为1764bp,该基因编码587个氨基酸。与已经报道的高等植物NiR序列同源性高达76%-88%,编码的氨基酸序列中含有一段典型的丝氨酸重复序列,基因登录号为GenBank:KC823634,并命名为MaNiR。同时利用基因编码的蛋白质序列与其它植物的亚硝酸还原酶基因构建了系统进化树,亚硝酸还原酶基因编码的氨基酸序列聚成了3支,分别为藻类、单子叶植物、双子叶植物。桑树MaNiR与多年生果树桦树、柑橘 、碧桃以及草莓的亲缘关系比较近。????

NiR primer was designed According to Morus notabilis genome database Scafford Morus02756. Guiyou62 cDNA was used as PCR template to get the full length .The full length of cDNA was 1764bp, encoding 587 amino acids . Sequencing analysis showed that it was 76% to88% identical with the sequences of other reported plants. the gene sequence is submitted to NCBI for login, the accession number is: Gen Bank: KC823634.we name as MaNiR.Using Muscle software to conduct the multiple comparison of NiR amino acid sequence and constructing the phylogenetic tree to conduct N-J clustering analysis. Nitrite reductase gene encoding amino acid sequences into three, respectively, algae, monocoty-ledon and dicotyledon.The genetic relationship between MaNiR and perennial fruit tree birch, citrus, prunus persica and strawberry is very close.
