流体细胞培养系统对三维培养的成骨细胞的影响
Effect of fluid flow cell culture system on osteoblasts seeded on 3D scaffolds
设计制作流体细胞培养系统并研究流体剪切力对三维培养成骨细胞黏附增殖的影响。方法:取SD乳鼠颅盖骨,采用组织块法获取成骨细胞,接种于部分脱蛋白天然牛骨支架,将实验组支架置于自制的细胞培养系统中,流速设置为0.3ml/min作用1d后增加流速至1ml/min, 对照组支架静置培养。4d和8d后收集实验组和对照组样本,SEM观察细胞在支架表面和内部的黏附、生长情况。SEM观察显示:流体剪切力作用下的细胞增殖分化较快,较早地向支架内部爬行生长而且细胞长轴明显沿加力方向延长。本实验成功制作了流体剪切力细胞培养系统并在体外证实流体剪切力有利于细胞在支架上黏附和增殖。
Objective To design and fabricate the fluid flow cell culture system, study the effect of fluid shear stress on the adhesion and proliferation of osteoblasts seeded on the 3D scaffold. Methods The third passage of osteoblasts harvested from the cranial bones of 2-3 day-old SD rat (2×106/100μl) was seeded on top to the partial deproteinized bones. Thereafter, the seeded scaffolds were cultured in the fluid flow cell culture system or under static condition for 4 and 8 days. The adhesion and proliferation of osteoblasts were evaluated by scanning electron microscopy(SEM). Results The SEM analysis showed a significant increase in cell proliferation for scaffolds that were subjected to fluid shear stress applied by the fluid flow cell culture system, and the axis of osteoblasts lengthened along the fluid flow direction. Conclusion This study designed and fabricated the fluid flow cell culture system, and demonstrated the fluid shear stress could enhance the adhesion and proliferation of osteoblasts seeded on 3D scaffolds.
