荧光法应用于杜仲提取物对补骨脂素血浆蛋白结合作用影响研究
Interaction between the Psoralen with Human Serum Albumin (HSA) and the Influence of the Coexisting on Eucommia extract on the Psoralen -HSA Binding System: by Fluorescence Studies
目的:应用荧光光谱法研究不同炮制时间杜仲提取物对补骨脂素血浆蛋白结合率的影响,为杜仲炮制时间合理化及杜仲补骨脂联合用药提供一定的科学依据。方法:将杜仲提取物与人血清蛋白定量混合,依次改变补骨脂素的浓度,设定激发波长280 nm,进行发射波长300-500 nm全波长荧光光谱扫描,记录杜仲提取物多成分共存体系下补骨脂素对人血清蛋白的荧光淬灭光谱。结果:补骨脂素与人血清蛋白的荧光淬灭过程为静态淬灭;2h和4h杜仲提取物共存体系中补骨脂素的血浆蛋白结合率明显降低,未炮制和1h杜仲提取物对补骨脂素蛋白结合作用无明显影响。结论:2h和4h炮制杜仲提取物可与补骨脂素产生竞争性蛋白结合作用,影响补骨脂素药效发挥,可为杜仲炮制及临床中药配伍提供借鉴。
objective: .In order to discuss the effects of the coexisting components from Eucommia extract (0h、1h、2h、4h processed respectively) on the Psoralen-HSA binding system, we carried out series fluorescence spectrometry studies of the Psoralen-HSA binding system in the presence of those components. Methods: The excitation wavelength was 280 nm,and the emission spectra were recorded from 300 to 500 nm. Fluorescence Spectroscopy was applied for the study of psoralen and human serum albumin coexisting system in presencing of eucommia extract. Results: Eucommia extract (2h、4h processed respectively) decreased the binding parameters Kb of the Psoralen-HSA system. The changes in the other binding parameters, such as Kq, Ksv, and Ka, were similar to those of Kb. Therefore, the participation of Eucommia extract (2h、4h processed respectively) may affect the interaction of the Psoralen-HSA system. Conclusion: The study will be used to guide Chinese medicine development, which are including Eucommia and Psoralen, be as reference of the compatibility of Psoralen and Eucommia in clinical use, provide ideas of the compatibility of traditional Chinese medicine research and references of pharmacokinetic study of compatibility of traditional Chinese medicine.
