高效液相色谱拆分两种β-受体阻断剂药物

建立了水溶液中β-受体阻断剂药物(普萘洛尔和美托洛尔)的固相萃取前处理方法和高效液相色谱荧光检测方法。使用Oasis和Supel两种HLB固相萃取柱进行样品前处理,水溶液中普萘洛尔和美托洛尔的回收率最高可达97.33%和96.08%。使用Chirobiotic T手性拆分柱,以甲醇:冰醋酸:三乙胺(100:0.01:0.01,v/v/v)为流动相,流速为0.8 mL/min,柱温20 ℃,可对两种β-受体阻断剂药物的手性异构体进行有效分离。该方法准确、简便、可靠,可用于水样中普萘洛尔和美托洛尔对映体的检测。
Solid-phase extraction (SPE) pre-treatment method and HPLC fluorescence detection method were established to detect aqueous solution of two β-receptor blocker drugs (propranolol and metoprolol). Using two HLB SPE columns (from Oasis and Supel company) for sample pre-treatment,the recovery rates of propranolol and metoprolol in aqueous solution reached 97.33% and 96.08%, respectively. The enantiomeric resolution of propranolol and metoprolol was studied using a teicoplanin based chiral stationary phase colume, with methanol: acetic acid: triethylamine (100: 0.01: 0.01, v/v/v) used as mobile phase at a flow rate of 0.8 mL/min at 20℃. Chiral isomers of two β-blocker drugs can be separated effectively. This method is accurate, simple and reliable, which can be used for propranolol and metoprolol enantiomer detection in water samples.
