罗格列酮抗高糖高胰岛素诱导心肌肥大作用研究
Effects of rosiglitazone in cardiac hypertrophy induced by high glucose and insulin
目的 研究罗格列酮(Rosiglitazone,RSG)在高糖高胰岛素(High glucose and insulin,HGI)诱导心肌肥大中的作用及机制。方法 利用乳鼠心肌细胞培养,以细胞表面积、蛋白含量和心钠肽mRNA表达为心肌肥大指标,观察RSG对HGI诱导心肌肥大的作用;利用硝酸还原法和ELISA法分别检测一氧化氮(Nitric oxide,NO)含量和内皮型一氧化氮合酶(Endothelial nitric oxide synthase,eNOS)的活性;Real-time PCR 和 Western blotting 方法分别检测mRNA和蛋白水平的表达。结果 HGI诱导心肌细胞肥大;RSG(0.01,0.1,1μmol/L)呈浓度依赖性地抑制HGI诱导的心肌细胞肥大;并明显上调HGI所导致的过氧化物酶体增殖物激活受体-γ(Peroxisome proliferator-activate receptor-γ,PPAR-γ)和eNOS mRNA表达的减少,同时增加PPAR-γ蛋白表达、eNOS活性以及NO含量(P<0.05)。PPAR-γ抑制剂GW9662以及NOS抑制剂L-硝基-精氨酸甲酯(L-NAME)可分别阻断RSG的上述作用。结论 RSG通过激活PPAR-γ受体,增加eNOS活性,促进NO释放,从而产生抗HGI诱导的心肌细胞肥大作用。
Objective To investigate the role of rosiglitazone (RSG) in high glucose and insulin (HGI)-induced cardiac hypertrophy and its mechanisms. Methods The cardiomyocyte hypertrophy responses were assayed by measuring the cell surface area, total protein content, atrial natriuretic peptide (ANP) mRNA expression in cultured neonatal rat cardiomyocytes. Nitric oxide (NO) concentration and endothelial nitric oxide synthase (eNOS) activity were determined by using the nitrate reduction and ELISA method, respectively. The mRNA and protein expressions were detected by real-time PCR and Western blotting, respectively. Results RSG(at 0.01, 0.1, 1 μmol/L) could inhibit the HGI-induced cardiomyocyte hypertrophy in a dose-dependent way. RSG significantly increased both mRNA and protein expressions of peroxisome proliferator-activate receptor-γ (PPAR-γ), elevated mRNA expressions and enzyme activity of eNOS, and raised NO concentration in cultured media. These effects of RSG could be abolished by GW9662, a PPAR-γ inhibitor, or NG-nitro-L-arginine-methyl ester(L-NAME), a NOS inhibitor, respectively. Conclusions RSG attenuates cardiomyocyte hypertrophy induced by high glucose and insulin, and the underlying mechanisms may be by activating PPAR-γ, increasing eNOS activity, thereby promoting NO release.
