四个miRNA在鸡性分化早期的性别差异表达谱分析

miRNA 是一类长度约为20~24 个核苷酸的小分子非编码RNA,它通过调控靶基因转录后的表达而在多种生物学途径中发挥作用。为探讨miRNA在性腺早期发育中的作用,本课题组在前期通过基因芯片方法筛选出了在E3.5~6.5 天雌、雄鸡胚中呈差异表达的若干个miRNAs,本研究即采用茎环状引物的定量PCR方法对其中4个miRNAs 进行表达谱分析。结果表明,miR-107、1767这2个miRNAs呈现前期雌性高表达后期雄性高表达的趋势,miR-2954的表达则是在E4.5d雄性中极显著高于雌性(P<0.01),miR-92在所检测的各时期(除E4.5d外)中的表达均为雄性极显著高于雌性 (P<0.01)。本文根据表达谱结果推测了miR-92可能涉及早期鸡胚性腺发育过程。
miRNA is a family of small, non-coding RNA molecules of about 20-24 nucleotides, which regulate many biological pathways by down-regulate target gene expression at the post-transcriptional level. In order to investigate the function of miRNA on the early development of chicken gonad., some sexually dimorphic miRNA expressions during E3.5~6.5 in chicken embryos had been found by miRNA chip in previous studies. In this study, expression patterns of 4 miRNAs which selected based on the early study were studied by stem-loop Quantitative PCR methods. The results showed miR-107 and 1767 were higher expression in female during early stage, but higher in male during later stage; miR-2954 was with significant higher expression in male at E4.5d(P<0.01), and miR-92 was expressed at significant higher level in male at all detected stages, except for E4.5d (P<0.01). Based on the expression results, it is speculated that miR-92 should have effected on the early process of gonad development in chicken embryo.
