低磷胁迫下杉木针叶蛋白的提取与表达差异分析

本文采用TCA-丙酮沉淀高速离心法,同时对杉木针叶蛋白质组双向电泳技术中的全细胞蛋白的提取、蛋白的裂解、电泳条件等环节进行了优化改良,获得了高重复性、高清晰度的杉木针叶响应低磷胁迫的差异蛋白图谱。在此基础上对低磷处理5d的杉木进行针叶蛋白提取。并使用ImageMaster 2D Platinum 7软件分析比较杉木针叶在对照与低磷处理下的蛋白表达谱。研究表明图谱中有59蛋白点存在显著差异可能与低磷胁迫有关。其中,上调表达点52个,下调表达点7个。

In order to obtain the protein profile with high repeatability and resolution under the condition of phosphorus deficiency, the high-speed centrifugation of TCA-acetone precipitation and the optimized electrophoresis including the extraction of whole-cell protein in Chinese fir needles, the protein cleavage and the condition of electrophoresis were used in this study. The protein extraction of fir needles under 5d low-phosphorus treatment was conducted, and the comparison of protein profile between control and the treatment samples were made by using ImageMaster 2D Platinum 7 software. The results showed that there were 59 protein points of low phosphorus treatment significantly different from the control sample, which included 52 upregulated and 7 downregulated points.
