二妙丸中三种生物碱成分在beagle犬体内的药代动力学研究
Simultaneous Determination of Berberine, Jatrorrhizine and Palmatine in Beagle Plasma by LC-MS/MS and its Application to Pharmacokinetic Study of Ermiao Wan
目的:建立高灵敏的测定血浆中小檗碱、巴马汀和药根碱浓度的LC-MS/MS法,并用于研究beagle犬灌服市售二妙丸后体内三种生物碱的药代动力学特征,DAS2.0软件计算药代动力学参数。方法:6只健康beagle犬,雌雄各半,分别单剂量灌服市售二妙丸制剂(相当于小檗碱25mg/kg)后,0~48小时间隔采集血样。结果:在本文建立方法下,小檗碱、药根碱和巴马汀的最低检测浓度为0.1,0.01和0.08ng/mL,血浆中小檗碱、药根碱和巴马汀的Cmax分别为(4.474±1.8)、(0.5083±0.2)和(0.8124±0.7)ng/mL,t1/2分别为(38.1±15.4)、(23.8±9.9)和(20.2±17.1)h,AUC(0-τ)分别为(37.4±3.9)、(3.75±1.2)和(3.01±0.9)ngoh/mL。结论:本方法准确可靠,灵敏度高,适用于极低的血药浓度分析,二妙丸中原小檗碱类成分在beagle犬体内吸收速率快、消除缓慢,可为二妙丸的临床药代动力学研究提供参考。
o establish an LC-MS/MS method for the simultaneous determination and plasma pharmacokinetic study of berberine, jatrorrhizine and palmatine in beagle dogs after oral administration of Ermiao wan. Six dogs (3 males and 3 females) were included in the study with a single orally dose of Ermiao wan equivalent of 25mg/kg berberine. Blood samples were collected into heparinized tubes in predetermined time intervals from 0 to 48 hours. The plasma concentrations of berberine, jatrorrhizine and palmatine were determined by the LC-MS/MS. The pharmacokinetic parameters were calculated by DAS2.0 software. The main pharmacokinetic parameters of berberine, jatrorrhizine and palmatine were found as follows: Cmax (4.474±1.8), (0.5083±0.2) and (0.8124±0.7) ng/mL, t1/2 (38.1±15.4), (23.8±9.9) and (20.2±17.1) h,AUC(0-τ) (37.4±3.9), (3.75±1.2) and (3.01±0.9) ng h/mL. The established LC-MS/MS method was sensitive and accurate for the simultaneous determination and pharmacokinetic study of berberine, jatrorrhizine and palmatine after a single oral administration of Ermiao wan in beagle dogs. The results indicated that the proberberines were absorbed quickly and eliminated slowly after oral administration. These may provide useful information for clinical pharmacokinetic study.
