补体活化对人外周血CD3+CD56+NKT细胞功能的负性调控作用
Negative regulation of human CD3+CD56+NKT-like cells by complement
目的 探讨补体活化对人外周血CD3+CD56+NKT细胞功能的影响。方法 密度梯度离心法分离PBMC,FCM检测CD3+CD56+NKT细胞补体受体及调节蛋白 的表达情况;免疫磁珠纯化法分离CD3+CD56+NKT细胞,分别加入正常血清和C3去除血清,比较CD3+CD56+NKT细胞功能受体CD69、NKG2D、CD158b和杀伤功能标志物CD107a以及细胞因子IFN-γ的表达情况,ELISA法检测细胞培养上清补体活化情况。结果 CD3+CD56+NKT细胞表达补体受体及调节蛋白;补体活化后不但下调兴奋性受体CD69、NKG2D,而且同时抑制了杀伤功能标志物CD107a以及细胞因子IFN-γ的分泌。结论 补体可能通过与补体受体结合负性调控CD3+CD56+NKT细胞的功能。
Objective: Study the effect of complement activation on the effector function of human CD3+CD56+NKT-like cells. Methods: PBMCs were isolated using Ficoll-Hypaque densitygradient centrifugation.The protein expression of complement receptors was confirmed by flow cytometry on CD3+CD56+gated cells. CD3+CD56+NKT-like cells were purified and cultured in the presence of normal serum(NS) or C3 depleted serum (C3-/-) at 37℃ 5% CO2 incubator for 24h, and the expression of surface molecules(CD69,NKG2D,CD158b,CD107a) and cytokine(IFN-γ) were measured by FCM. The level of C3 fragments (i.e. C3a, iC3b) in the cell culture supernants was measured by ELISA. Results: human CD3+CD56+NKT-like cells are able to express most complement receptors and membrane regulators. Complement activation inhibitted the expression of CD69, NKG2D ,CD107a and the cytokine of IFN-γ on CD3+CD56+NKT-like cells.Conclusion:complement activation represents a novel mechanism for negative regulation of the CD3+CD56+NKT-like cells activation and effector function, the regulation could be mediated by complement receptor-ligand interaction
