一株腈水合酶产生菌的驯化、保存与活化

本文研究了腈水合酶产生菌株Rhodococcus sp.JSHM-1的驯化、保存与活化条件。实验结果表明,用丙烯腈对其进行梯度驯化,当丙烯腈的浓度达到8 g/L时,驯化效果最明显,菌株的酶活力由原来的476.8U提高到962.4U。在菌种的保存过程中,加入浓度为4g/L的丙烯腈时菌种的酶活力保持的最好,仍能达到927.5U。在对老化的菌种进行活化时,固体培养丙烯腈活化的方法优于液体培养丙烯腈活化的方法;采用固体培养丙烯腈活化的方法,丙烯腈的浓度为4g/L时,菌种活化的效果最好,菌体的酶活由老化后的596.2U提高到活化后的971.6U。
he conditions of domestication, preservation and activation on a nitrile hydratase producing strain Rhodococcus sp.JSHM-1 were studied in this paper. The results show that domesticating the strain gradually using acrylonnitrile, when the concentration of acrylonnitrile accounted to 8 g/L, the result of domesticating was the most obvious, that was the enzyme activity raised from 476.8U to 962.4U. During the period of preservation, the enzyme activity of the strain was kept best when the concentration of acrylonnitrile was 4g/L, which was 927.5U. When activated the aged strain, the method of solid culture was better than fluid culture, and the enzyme activity under the solid culture method as adding 4g/L acrylonnitrile could increase to 971.6U although it was only 596.2U before activating.
