目的 探讨n-3多不饱和脂肪酸对NOD小鼠T细胞免疫学功能的影响。方法 取野生型Balb/c小鼠和发病NOD小鼠脾脏细胞,经磁珠分选后获得小鼠CD4+ T细胞后,将其分为4组,野生型小鼠为Wild type组,NOD鼠分为未处理组(Control)、DHA处理组、EPA处理组。经DHA、EPA处理24h,PMA和Ionomycin刺激活化后,采用CCK-8法检测T细胞增殖、采用流式细胞仪检测Th1/Th2极化、采用ELISA法检测T细胞细胞因子的分泌水平变化。结果 DHA、EPA对T细胞增殖具有显著抑制作用;促进NOD小鼠Th2细胞分化,抑制Th1细胞分化;经ELISA检测,DHA和EPA能够抑制T细胞IL-6、IL-17的分泌。结论 n-3多不饱和脂肪酸通过抑制T细胞增殖,平衡Th1/Th2比例和抑制IL-6、IL-17的分泌对NOD小鼠的T淋巴细胞起到免疫抑制作用。

Objective Study the effects of n-3 polyunsaturated fatty acid PUFAs on immunological functions of T cells in NOD mice. Methods T cells from Balb/c and NOD mice were treated with 50?M EPA and DHA, then the viability of T cells was tested through CCK-8 kit, the differentiation of Th cells was tested and analysed by flow cytometer, and the secretion of inflammatory cytokines were tested by ELISA Kits. Results EPA and DHA could inhibit the viability of T cells from NOD mice, suppress the na?ve T cells differentiate into Th1 cells,balance the ratio of Th1 and Th2, and downregulate the secretion of IL-6 and IL-17. Conclusion n-3 PUFAs could suppress some immune functions of T cells through inhibiting their viability, Th1 polarization and inflammation cytokines secretion.
