探讨溶栓及抗凝药物对内皮细胞内源性纤溶和抗凝功能的影响。将药物按一定浓度稀释,之后与凝血酶预处理过的人肺动脉内皮细胞(Human Pulmonary Arterial Endothelial Cells,HPAECs)共同孵育,收集培养上清并应用ELISA方法检测其中尿激酶型纤溶酶原激活物受体(urokinase-type plasminogen activator receptor,u-PAR),纤溶酶原激活物抑制物-1(plasminogen activator inhibitor-1,PAI-1)及组织因子(tissue factor,TF)的含量。与凝血酶组相比,重组尿激酶原(Pro-urokinase)组u-PAR的含量显著增加(0.51±0.04 VS 0.37±0.01);重组尿激酶原组和组织型纤溶酶原激活物组PAI-1的含量显著下降(53.4025±3.09,44.1750±1.88 VS 107.9963±5.95);组织型纤溶酶原激活物(recombinant tissue-type plasminogen activator,rt-PA)组和低分子肝素(Low-molecular-weight Heparin Calcium,LMWH)组TF的含量显著下降(116.6313±7.49,156.7563±11.07 VS 231.4063±20.42)。重组尿激酶原、重组组织型纤溶酶原激活物及低分子肝素对人肺动脉内皮细胞的内源性纤溶和抗凝功能的影响不同。这可能会影响血栓性疾病治疗中临床药物的选择。

o study The effect of fibrinolytic or anticoagulant drugs on the fibrinolytic function of HPAECs. Methods When the HPAECs reached confluence, starvate for 12 h, and incubate the cells with different drugs for 12 h. Cells were classified into 5 group according to different drug: control group, thrombin(1 IU/ml) group; thrombin (1 IU/ml) and pro-urokinase (150 IU/ml) group, thrombin (1 IU/ml) and rt-PA (150 IU/ml) group and thrombin (1 IU/ml) and LMWH (10 IU/ml) group. After incubation, collect the medium, and detect the content of u-PAR, PAI-1 and TF. Results Compared to thromin group, the content of u-PAR in pro-urokinase group was statistically increased (0.51±0.04 VS 0.37±0.01); The content of PAI-1 in Pro-urokinase group and rt-PA group were statistically decreased(53.4025±3.09,44.1750±1.88 VS 107.9963±5.95); The content of TF in rt-PA group and LMWH group were statistically decreased(116.6313±7.49,156.7563±11.07 VS 231.4063±20.42). Conclusion Pro-urokinase, rt-PA and LMWH have direct and different effect on the fibrinolytic and anti-coagulant function of HPAECs, which may be taken into consider in different thromboembolic diseases.
