RNA干扰靶向沉默Ku70对人肺腺癌耐药细胞细胞周期与凋亡的影响
Effects of silencing of Ku70 by siRNA on Cell Cycle and Apoptosis of human lung adenocarcinoma cell
目的 采用RNA干扰靶向沉默Ku70基因,观察抑制Ku70对人肺腺癌耐药细胞(A549DDP)的细胞周期、凋亡的影响。方法 首先采用RT-PCR、Western Blot方法比较Ku70mRNA及蛋白在人肺腺癌亲代(A549)与耐药细胞系(A549DDP)的表达水平;将靶向Ku70siRNA质粒载体转染至A549DDP细胞后,再次检测其Ku70的mRNA、蛋白水平;应用分光光度法、流式细胞仪等多种方法,观察靶向沉默Ku70后A549DDP细胞在顺铂作用下的细胞形态学、Caspase-3活性以及凋亡相关指标的变化。结果 1.Ku70mRNA、蛋白在人耐药肺腺癌细胞表达水平显著高于其亲代非耐药人肺腺癌细胞;2.与未导入Ku70siRNA及导入非特异性siRNA的人肺腺癌耐药细胞相比较,靶向导入Ku70siRNA后A549DDP细胞的Ku70的mRNA、蛋白表达水平下调;在相同浓度的顺铂作用下,抑制Ku70的耐药细胞凋亡率增加,细胞周期阻滞于S期,线粒体膜电位下降,Caspase-3活性增加,细胞形态学呈凋亡增加的改变。结论 Ku70mRNA在人耐药肺腺癌细胞呈现高表达,提示Ku70参与肺腺癌耐药的形成;靶向Ku70siRNA导入人肺腺癌耐药细胞可特异性下调Ku70的表达,促进肺腺癌耐药细胞凋亡,故Ku70可能成为临床监测肿瘤耐药性变化的标志物,逆转肺癌耐药的一个新的分子靶点。
Objective In this paper ,to explore the role of Ku70 in the regulation of drug-resistance in lung cancer, we examined the changes of cell cycle and apoptosis of A549DDP by silencing Ku70 using small interfering RNA (SiRNA) . Methods Expression of Ku70mRNA and Ku70 protein were measured in parental A549 and A549DDP by RT-PCR and Western Blot. A549DDP was transfected with Ku70 siRNA targeting Ku70. The changes of the inhibition rate of cell morphology,cell cycle , apoptosis and caspase-3 activity were observed by flow cytometry and spectrophotometer.Results 1.The Ku70mRNA and protein expression were higher in A549DDP cells compared with those in A549 cells; 2.The expression of Ku70 mRNA and protein in A549DDP were downregulated after the siRNA plasmid transfected. With the equal concentrations of DDP the apoptosis rate increased;mitochondrial membrane potential declined;cell cycle arrested at S phase; and activity of caspase-3 increased ;more changes of apoptotic cells in morphology in Ku70 silencing A549. Conclusions Our data provide the functional study of Ku70 as an apoptosis and drug-resistance modulator , which show Ku70 might be involved in the development of drug resistance of lung cancer. Ku70 siRNA downregulated the expression of Ku70 and promted the apoptosis of A549DDP .Therefore, Ku70 may be the marker of resistance to chemotherapy for tumor and a new target that reverses the chemotherapy-resistant of lung cancer.
