血管紧张素-(1-7)对腹主动脉缩窄大鼠血管内皮功能的影响
Effects of Angiotensin-(1-7) on Endothelial Dysfunction in Rats with Coarctation Induced Hypertension
摘 要: 【目的】 探讨血管紧张素-(1-7)[Ang-(1-7)]对腹主动脉缩窄性高血压大鼠血管内皮功能的影响。【方法】 采用腹主动脉缩窄术制成腹主动脉缩窄性高血压大鼠模型,45只SD大鼠随机分为假手术组、模型对照组、Ang-(1-7)治疗组(25μg.kg-1.h-1,持续静脉给药)。4周后检测大鼠颈动脉血压、血浆NO2-/NO3-水平,胸主动脉环对乙酰胆碱(Ach)和硝普钠的舒张反应,并采用免疫印迹方法检测主动脉内皮型一氧化氮合酶(endothelial NOS,eNOS)和诱生型一氧化氮合酶(inducible NOS,iNOS)的表达。【结果】与假手术组比较,模型对照组颈动脉血压明显升高(P<0.01)、血浆NO2-/NO3-水平也明显增高(94.7 μmol/L vs 40.1 μmol/L,P<0.01),Ach引起的血管环舒张百分比下降(39.4% vs 73.8%, P<0.01),主动脉eNOS表达降低、iNOS表达增加。与模型对照组比较,Ang-(1-7)治疗未导致颈动脉血压、血浆NO2-/NO3-水平明显改变,但Ach引起的血管环舒张百分比增加至57.4%,并改善了主动脉eNOS和iNOS的异常表达。 【结论】 外源性Ang-(1-7)可改善腹主动脉缩窄性高血压大鼠的血管内皮功能,这种作用可能与调节一氧化氮合酶的表达有关。
【Objective】 To investigate the effects of angiotensin-(1-7) on endothelial dysfunction in rats with coarctation induced hypertension. 【Methods】 Hypertensive rat models was induced by constriction of abdominal aorta. Forty-five male Sprague Dawley rats were randomized to sham-operated group, model control group and angiotensin-(1-7) group. They were treated with intravenous infusion of angiotensin-(1-7) (25 microgram/kg per hour) or saline after operation. The levels of blood pressure and palsma NO were determined at 4 weeks after operation. Acetylcholine(Ach)-induced vasodilation was measured in rat thoracic aorta. Western blot analysis was performed to valuate the eNOS and iNOS protein expression of aorta. 【Results】 The levels of blood pressure and plasma NO in model control group were significantly higher than those of sham-operated group. Compared with sham-operated group ,Ach-induced vasodilation was decreased,the expression of eNOS was decreased and the expression of iNOS was increased in model control group.The levels of blood pressure and plasma NO in ang-(1-7) group were not changed compared with those in model control group. However, angitensin-(1-7) improved the vasdilation response of aorta to Ach,and normalized the expression of eNOS and iNOS. 【Conclusion】 Angiotensin-(1-7) can improve vascular indothelial dysfunction by promoting eNOS protein expression in rats with coarctation induced hypertension.
