为在细胞以及分子水平研究家禽心肌的生物学特性,建立分离以及培养鸡胚心肌细胞的方法,以此提供研究心脏分化以及心血管疾病病理过程的体外模型。建立9-10天鸡胚分离培养的方法,在鸡胚分离三天后,通过镜下观察细胞形态,并用抗α- 横纹肌肌动蛋白免疫组织化学染色方法鉴定培养的心肌细胞。结果观察到心肌细胞呈梭形、星形、不规则形,95%的抗α- 横纹肌肌动蛋白免疫组织化学染色阳性的心肌细胞成片同步收缩,证明我们已经建立了良好的分离培养鸡胚心肌细胞的方法。
Isolation and characterization of chicken embryonic cardiomyocytes are critical for understanding on their biological properties. Cultured the chicken embryonic cardiomyocytes is one of key technologies to do some research about cardiac development, homeostasis and cardiovascular diseases. Here we described a method for the isolation and identification in vitro culture of chicken embryonic cardiomyocytes. Isolated cardiomyocytes were identified by cellular morphology and immunocytochemistry. The results showed that the cultured cardiomyocytes from the chicken embryos exhibited synchronous contraction and typical morphology, and stained positive for α- sarcomeric actin viewed under an inverted microscope. In conclusion, we provide a protocol for the reliable method of isolation and cultivation of chicken embryonic cardiomyocytes may allow more detailed analysis of their properties, and provide a valuable model for studying pathological processes including pulmonary hypertension, ascites and cardiovascular diseases.
