去分化间充质干细胞体内成骨再分化潜能的实验研究

目的:探究人骨髓源的去分化间充质干细胞(De-MSCs)在体内成骨再分化的能力。 方法:将成骨诱导培养基重悬的De-MSCs与间充质干细胞(MSCs)以相同浓度种植在胶原模块上,并置于培养箱中培养3小时后,将负载细胞的胶原模块植入重症联合免疫缺陷(SCID)小鼠皮下进行体内异位成骨实验。植入7天时,取出植入的胶原模块进行碱性磷酸酶(ALP)活性检测和荧光定量PCR(RT-PCR)检测成骨基因的表达;植入30天时,取出胶原模块进行HE染色和Col II免疫组织化学染色。结果:ALP活性检测结果显示De-MSCs再成骨组的ALP含量明显高于MSCs成骨组(P<0.01)。RT-PCR结果显示De-MSCs组的成骨相关基因(BMP2,Runx2,Osterix和DLX5)的表达水平均明显高于MSCs组(P<0.01)。HE染色结果显示De-MSCs组与MSCs组均可见明显异位新骨形成,而免疫组化染色结果可见De-MSCs组的骨组织中成骨细胞胞膜的II型胶原的阳性表达率高于MSCs组。结论:De-MSCs较MSCs在体内具有更强的成骨分化潜能,De-MSCs可作为更优质的种子细胞应用于组织工程和再生医学。
Objective: To explore the osteogenic redifferentiation potential of de-differentiated MSCs (De-MSCs) in vivo. Methods: De-MSCs and MSCs were resuspended in osteogenic induction medium and seeded in collagen scaffolds with the same concentration. The scaffolds with cells were subcutaneously implanted into SCID mice. After implantation for 7 days, the collagen scaffolds were collected and ALP activity assay and RT-PCR assay were performed to detect the expression of osteogenic related genes. After implantation for 30 days, the collagen scaffolds were collected and HE staining and immunohistochemistry staining were performed. Results: The results of ALP activities and expression levels of osteogenic related genes (BMP2, Runx2, Osterix and DLX5) in De-MSCs group were obviously higher than those in MSCs group. The HE staining of De-MSCs group and MSCs group showed ectopic new bone formation. The results of immunohistochemistry staining showed the positive expression rate of collagen type II of osteoblast cell membrane in De-MSCs group was higher than that in MSCs group. Conclusions: Compared with MSCs, De-MSCs have higher osteogenic differentiation efficiency in vivo. De-MSCs could be an alternative potential seed cells in tissue engineering and regenerative medicine.
