摘要

内源激素作为调控种子萌发与休眠的主要内因一直备受关注,但对高寒植物种子内源激素调控种子休眠与萌发对环境条件适应性的研究一直缺乏普遍接受的理论或假说。本研究以青藏高原东部高寒草甸退化草地常见种露蕊乌头(Aconitum gymnandrum)种子为研究对象,分析不同海拔的种子在常温和冷藏条件下内源激素(ZT、GA、IAA、ABA)含量的动态变化及其对种子休眠与萌发的影响。结果表明:1)低温冷藏有利于维持露蕊乌头种子休眠状态,高海拔露蕊乌头种子萌发对温度响应更加敏感,无论何种储藏条件种子内促进类激素ZT、GA、IAA含量及ZT/ABA、GA/ABA、IAA/ABA比值都要高于低海拔种子,而抑制类激素ABA含量则要低于低海拔种子,然而种子萌发率仅在常温储藏下显著高于低海拔种子;2)两种储藏条件下,露蕊乌头种子均维持较高的休眠状态(最高萌发率仅为37.33%),常温储藏条件下种子萌发率显著高于4℃冷藏条件下,但种子中内源激素含量及其与ABA的比值并未表现出明显的变化趋势;3)IAA是露蕊乌头种子休眠释放必不可少的内源激素,其含量正向调节种子萌发,随储藏时间的延长,只有IAA含量与种子萌发率显著正相关。

Abstract

Seed germination and dormancy are affected endogenous hormones have been gained great attention. few studies on the regulation of endogenous hormones on seed dormancy and germination of alpine plants and the adaptation of their contents variation to environmental cues were focused on. In this study, the seeds of Aconitum gymnandrum (Ranunculaceae), a common species from degraded grassland in the alpine meadow on the eastern Qinghai-Tibet Plateau, was used to explore the influences of endogenous hormones contents of seeds on seed dormancy and germination. Contents of endogenous hormones (ZT, GA, IAA, ABA) and germination percentage of seeds were determined under different storage conditions, storage and altitudes. The research results showed as follows: 1) Low temperature refrigeration is beneficial to maintain the dormancy of the seeds of Aconitum gymnandrum, which high altitude were more sensitive to temperature. Regardless of the storage conditions, the levels of ZT, GA, IAA and ZT/ABA, GA/ABA, and IAA/ABA in the seeds are higher than those in low-altitude seeds. on the opposite, the contents of inhibitor, ABA, were lower, however, only under room temperature storage the germination ratio is significantly higher than that in low-altitude seeds; 2)The dormancy levels under two different storage conditions were high (the highest germination is 37.33%, the seed germination is significantly higher under room temperature, but the endogenous hormone contents and their ratios with ABA did not show obvious change trend. So dormancy release is not caused by the abosolutely increase of the promoters or the decrease of the inhibitors; 3) IAA might be essential to break seeds dormancy and promote seeds germination of Aconitum gymnandrum. the percent germination was only positively correlated with the content of IAA with storage time.
