猪链球菌2型缺失菌株ΔNisKR免疫保护力的研究
Immune effect of a ?NisKR mutant strain of Streptococcus suis 2 for potential use in a live attenuated vaccine
猪链球菌是一种猪源性病原菌,也可以引起人的急性系统性感染,其中猪链球菌2型的流行性最强,给人类及养猪产业带来严重的威胁。为了更有效的控制猪链球菌2型病的发生及传播,研发具有活性的候选疫苗是一种有效途径。本研究呈现了构建自猪链球菌2型的基因缺失候选疫苗,ΔNisKR。通过免疫实验发现,被缺失株ΔNisKR免疫过的小鼠对抗致死剂量猪链球菌2型感染能力明显提高,结果显示ΔNisKR为小鼠提供了很强的免疫保护力。通过免疫实验发现缺失株ΔNisKR引起了小鼠体内很强的体液免疫,高效诱导了抗体 IgG1和IgG2亚型的产生。在全血实验和被动免疫实验中,抗突变株血清提供了50%的免疫保护率。而且在淋巴细胞增生实验中一个明显的增殖T细胞免疫反应被注意到。通过一系列的免疫实验,证明了ΔNisKR能够为宿主提供显著的免疫保护力,同时也阐述了基因缺失弱毒苗激发免疫保护的机制,为应对猪链球菌2型的感染提供了一个有效的策略。
Streptococcus suis (S. suis) is an important zoonotic pathogen causing severe systemic infection in humans, especially serotype 2, causes significant economic losses to the swine industry and threats to human health worldwide. In order to more effectively controlling the occurrence and spread of S. suis 2 disease, development of the a live attenuated candidate vaccine is an effective way. In this study, ΔNisKR derived from SC19 is studied as a live attenuated vaccine, which provided significantly greater protective efficacy against challenge with a lethal dose of S. suis 2 in a mouse model compared with the blank or negative control groups. The results revealed that the ΔNisKR mutant elicited a strong humoral antibody response; both IgG1 and IgG2 subclasses were induced. Moreover, the antisera against the mutant were bactericidal in a whole blood assay and provided 50% protective efficacy against S. suis 2 infection in a passive immunization experiment. In addition, a significant proliferative T-cell immune response was noted in a lymphocyte proliferation assay. Performing numerous immunological assays for determination of immunogenicities, we demonstrated that the mutant immunization, which provided significant protective immunity, would offer insight into the underlying immune mechanisms of a bacterial live attenuated vaccine and provide a novel strategy against S. suis 2 infection.
