家蚕Bm050524基因的表达、纯化及转录分析
Expression, purification and transcription analysis of a novel gene Bm050524 from silkworm, Bombyx mori
家蚕Bm50524基因是一个全新的基因,迄今为止未见相关文献报道对此基因进行研究。Bm50524基因开放阅读框为483bp,编码160个氨基酸的未知蛋白,预测分子量为18.56 kDa,等电点为5.11。基因结构分析表明Bm50524基因含有三个外显子。在E.coli BL21(DE3)中表达融合蛋白His- Bm50524,发现融合蛋白以包涵体形式存在。利用亲和层析的方法纯化融合蛋白,制备多克隆抗体。提取家蚕五龄幼虫七个组织总蛋白,荧光Western blotting分析发现Bm050524蛋白在家蚕七个组织中都有表达。提取家蚕五龄幼虫九个组织的总RNA,荧光定量PCR分析表明Bm050524基因在家蚕五龄幼虫头部转录水平较高。我们的实验为深入研究Bm050524基因功能奠定了基础。
he gene Bm050524 is a novel gene from silkworm, Bombyx mori . Up to now, there is no literature reporting about any characters about this gene. Bm050524 contains an open reading frame(ORF) of 483bp nucleotides , and encodes 160 amino acids with the predicted molecular weight of 18.56 kDa. We also took genome alignment between Bm050524 cDNA and silkworm genome, and the results showed that this gene includes three exons. The recombinant protein of His-Bm050524 was expressed in E.coli BL21 (DE3) and existed as inclusion body. After purification by nickel affinity column chromatography, polyclonal antibody was obtained. Total protein of seven tissues of the fifth instar larvae were extracted and analyzed by Fluorescent Western blotting. The results showed that Bm050524 widely existed in the tissues of the fifth instar larvae. Total RNA of nine tissues of the fifth instar larvae (Bombyx.mori) were extracted for RT-PCR experiment. RT-PCR demonstrated there were high transcriptional level head compared to other tissues. These results laid a good foundation of further studies of Bm050524.
