响应面法优化棕壤茶园专用解磷微生物菌剂制备工艺
Optimization of Preparation Technology for Brown Tea Garden Special Phosphate Solubilizing Microorganism Agent by Response Surface Methodology
(目的)为了探究山东茶园专用解磷微生物菌剂的制备工艺,(方法)本试验采用响应面法对棕壤茶园专用解磷微生物菌剂的制备工艺进行优化选择。(结果)试验通过Plackett-Burman设计获得了影响茶园微生物菌剂制备的三个关键因素:水与载体比值(X1)、发酵温度(X2)、烘干温度(X3)。然后利用最陡爬坡试验、中心组合试验以及响应面分析法,建立了以菌剂有效活菌数为响应值的二次回归模型。该模型预测最佳制备工艺条件为:水/载体(体积比)=1.8,菌液接种量为载体量的12.5%,发酵温度和烘干温度分别为30℃和41℃。采用上述优化工艺条件制备的菌剂,其有效活菌数与模型预测的最大值基本相符,说明预测模型准确可靠。
Response surface methodology was used in order to optimize the preparation technology for Brown tea garden special phosphate solubilizing microorganism agent. Three key factors that affect the preparation technology for tea microbial agent were got by Plackett-Burman design, including ratio of water and carrier (X1)、fermentation temperature (X2) and dry temperature(X3). A Quadratic regression equation model was established according to steepest ascent experiment, central composite design and response surface methodology , which the response value was the effective number of living bacteria in agents. The model predicted that the maximum number of the effective living bacterium was 5.01×1010 and the optimal preparation conditions were as follow: ratio of water and carrier was 1.8, the amount of bacterial inoculum was 12.5% of carrier, the temperature of fermentation and drying were 30℃ and 41 ℃ respectively. The effective number of living bacteria in agents that prepared using the above optimization process conditions which basically consistent with the maximum predicted value by the model. These results suggested that the predicted model was reliable.?????
