龙牙楤木皂苷IV对心肌细胞缺氧/复氧损伤的保护作用
Protective effects of Tarasaponin IV on Hypoxia/Reoxygenation injury in H9c2 Cardiocytes
目的:观察龙牙楤木皂苷IV对缺氧复氧诱导H9c2心肌细胞损伤的保护作用。方法:实验分为5组:空白对照组(C组);缺氧/复氧模型组(H/R组);缺氧/复氧模型+龙牙楤木皂苷IV低(12.5μg/ml)、中(25μg/ml)、高(50μg/ml)剂量组。利用缺氧/复氧(H/R)的方法诱导体外培养的H9c2心肌细胞损伤来模拟临床上心肌缺血再灌注损伤,观察龙牙楤木皂苷IV对缺氧复氧所致的H9c2心肌细胞损伤的保护作用。MTT法检测细胞的存活率; Hoechst33342/PI荧光染色法观察龙牙楤木皂苷IV对缺氧复氧诱导心肌细胞凋亡的影响以及JC-1荧光探针法检测H9c2心肌细胞线粒体膜电位的变化。结果:与空白对照组相比,H/R组细胞存活率显著降低(P< 0.01)。与H/R组相比,龙牙楤木皂苷IV各剂量组细胞存活率和线粒体膜电位明显升高(P< 0.05),并且显著降低心肌细胞凋亡率(P< 0.05)。结论:龙牙楤木皂苷IV对心肌细胞缺氧复氧损伤有明显的保护作用,呈剂量依赖性。其作用机制可能与降低心肌细胞凋亡,升高线粒体膜电位有关。
Objective:To investigate protective effects of Tarasaponin IV on H9c2 cardiac myocytes exposed to simulated hypoxia- reoxygenation. Methods: This experiment Were divided into five groups: normal control group (group C); hypoxia / reoxygenation model group (H/R group); hypoxia / reoxygenation model + Tarasaponin IV low (12.5μg/ml), medium (25μg/ml), high (50μg/ml) dose group . Use of cardiac H9c2 cells cultured in vitro, using hypoxia / reoxygenation (H/R) method of myocardial cell injury induced H9c2 to simulate the clinical myocardial ischemia-reperfusion injury, observe Tarasaponin IV of the hypoxia protective effects of H9c2 myocardial cell injury. Compared with the H/R group, cell viability and mitochondrial membrane potential was significantly increased(P<0.05) in Tarasaponin IV group, and significantly reduce myocardial cell apoptosis(P<0.05).Conclusion: Tarasaponin IV on myocardial hypoxia reoxygenation injury has a significant protective effect in a dose-dependent manner. The mechanism may be associated with decreased myocardial apoptosis, increased mitochondrial membrane potential.
