腺样囊性癌中自噬相关基因Beclin-1的表达及其与预后的关系

目的 自噬是胞浆内容物和细胞器在溶酶体内降解是营养和能量再利用的过程，与肿瘤的发生发展密不可分。但自噬在唾液腺腺样囊性癌各阶段以及患者预后中的作用尚不明确。本研究旨在通过检测腺样囊性癌组织中自噬相关基因Beclin-1的表达及临床意义。方法 采用免疫组织化学法检测89例腺样囊性癌组织中Beclin-1的表达情况,并分析其与临床特征以及预后的关系。结果 本组唾液腺腺样囊性癌组织中Beclin-1强阳性率为82.0% (73/89)，明显低于癌旁正常腺体（96.6%，P〈0.05）。Beclin-1的表达与肿瘤的病理分型密切相关（P=0.024）。经单因素和多因素分析,Beclin-1强阳性表达与良好的预后呈正相关（P〈0.05）结论 在唾液腺腺样囊性癌中Beclin-1的弱表达可能与肿瘤的发生有关，并且对腺样囊性癌患者具有独立的预后价值。
his study investigated the expression of autophagy gene Beclin-1 in human salivary adenoid cystic carcinoma (ACC) and their relationship with clinical pathological features. Immunohistochemistry and realtime-PCR were used to analysis their expression in ACC tissue samples. The results showed that significantly lower high-expression rate of this protein was found among ACC samples. Furthermore, Beclin-1 expression level was significantly correlated with histological pattern. Most importantly, univariate and multivariate survival analysis suggested that protein expression of Beclin-1 in cancer cells were independent prognostic indicators for ACC.
