采用SSR和SRAP分子标记构建葡萄分子遗传图谱
SSR AND SRAP MARKER BASED LINKAGE MAP OF Vitis vinifera L.
以葡萄品种'87-1'为母本,优良品系'9-22'为父本,杂交创建了F1群体。从该群体中随机选用149个单株组成作图群体,采用SSR和SRAP分子标记分析,并运用Joinmap3.0软件进行连锁分析,分别构建了母本'87-1'和父本'9-22'的分子遗传图谱以及双亲的共有图谱。其中,'87-1'的遗传图谱包括21个连锁群,包含163个标记位点,覆盖总图距1272.9cM,位点间平均遗传距离为8.9cM;'9-22'的遗传图谱包括20个连锁群,含有158个标记位点,覆盖总图距1267.4 cM,位点间平均遗传距离为9.1cM。共有图谱包括20个连锁群和一个二连体,包含217个标记,图谱总长为1537.1cM,标记间平均距离为7.8cM,连锁群的平均长度为69.8 cM。图谱覆盖了葡萄基因组的19条染色体,该图谱的构建为今后进一步开展相关性状的QTL定位和分子标记辅助选择等研究奠定了基础。
n F1 population was created by the cross '87-1'×'9-22'. The female parent 87-1 was an extremely early maturing cultivar with strong flavor. The male parent was an excellent breeding line producing large berries maturing late. 149 individuals were randomly chosen as the mapping population. Molecular genetic map for each parent as well as the consensus map were constructed using simple sequence repeat(SSR)and Sequence-Related Amplified Polymorphism(SRAP)marker by software JoinMap 3.0. The 87-1 map covers a total length of 1272.9 cM distributed in 21 linkage groups, and consists of 163 molecular markers with an average distance between adjacent markers of 8.9 cM. The 9-22 map covers a total length of 1267.4 cM distributed in 20 linkage groups, and consists of 158 molecular markers with an average distance between adjacent markers of 9.1 cM. The consensus map covers a total length of 1537.1 cM distributed in 21 linkage groups and one doublet, and consists of 217 molecular markers with an average distance between adjacent markers of 7.8 cM. The length of linkage group is 69.8 cM on average. The maps has covered the 19 chromosomes of Vitis genome, and can lay a solid foundation for further studies such as QTL mapping of correlated traits and marker assisted selection.
