姜黄素对鬼臼毒素细胞毒性的保护作用
Protective effects of curcumin against the cytotoxicity of Podophyllotoxin
目的 研究姜黄素对鬼臼毒素细胞毒性的保护作用及减毒机制。方法 选取VERO细胞,用MTT法检测鬼臼毒素的细胞毒作用,FCM检测细胞周期和细胞膜电位,用激光共聚焦显微镜观察微管的形成。结果 姜黄素可显著减轻鬼臼毒素的细胞毒作用,可缓解鬼臼毒素引起的G2/M期阻滞、细胞膜电位改变和对微管形成的影响。结论 姜黄素对鬼臼毒素的细胞毒性具有保护作用,其保护机制可能与其竞争性结合微管蛋白上的鬼臼毒素结合位点有关。
Objective To demonstrate the protective effect and mechanisms of curcumin administrastion against the cytotoxicity of podophyllotoxin. Methods Protective effects of curcumin against podophyllotoxin poisoning were examined in VERO cells. Cell toxicity was measured by MTT assay. Cell cycle and membrane potential were determined by flow cytometry. Microtubules were observed by confocal laser scanning microscope. Results Curcumin could attenuate podophyllotoxin-induced toxicity in VERO cells, recover podophyllotoxin-induced G2/M arrest, membrane potential change and the disturbance of microtubule structures. Conclusion Curcumin had protective effects against the podophyllotoxin induced cytotoxicity. The mechanisms might be associated with the competitive binding to tubulin.
