目的:通过免疫组化法观察复方浙贝颗粒联合阿霉素对K562/A02移植瘤细胞耐药相关酶GST、Topo-II表达影响。方法:将K562/A02细胞接种BALB/c-nu裸小鼠腋前皮下构建MDR移植瘤动物模型,给予复方浙贝颗粒灌胃联合阿霉素腹腔注射,实验结束后处死小鼠,剥离肿瘤,将瘤块组织制成切片,通过免疫组化结合Image ProPlus 6.0免疫组化彩色图象分析,观察复方浙贝颗粒联合阿霉素对K562/A02移植瘤细胞耐药相关酶GST、Topo-II表达的影响。结果:与阿霉素组比较,CZBG各剂量组移植瘤组织细胞相关酶GSH、Topo-II的IOD值明显降低(p<0.05)。结论:CZBG能协同阿霉素抑制K562/A02移植瘤组织细胞耐药相关酶GSH,TopoII的高表达。

Objective:To investigate the drug-combination of Compound Zhe Bei Granule (CZBG) and doxorubicin effects on the expression of GST and Topo-II in K562/A02 cell line multidrug resistance tumor xenografts in mice. Method:Tumor xenografts model were established by injecting the multidrug resistance cell line K562/A02 in the axillary flank of BALB/c-nu-nu mice. Drug-comgbination of CZBG intragastric administration and doxorubicin intraperitoneal injection (i.p.) were given to the BALB/c-nu nude mice. The tumor xenografts were made into slice after the dissection, and the expression ofGST and Topo-II in K562/A02 tumor xenografts in mice were investigated in immunohistochemical techniques. the integral optical density (IOD) of GST and Topo-II in K562/A02 tumor xenografts were measured by Image ProPlus 6.0. Results:Compared with the single treatment of doxorubicin i.p group., the groups of the combination of the doxorubicin and CZBG with dosage classified by three types(high, middle, low) can decrease IOD of GSH and Topo-II in K562/A02 tumor xenografts with statistical significance(p<0.05).Conclusion:The combination of CZBG and doxorubicin can decrease the expression of GST and Topo-II in K562/A02 multidrug resistance tumor xenografts in mice.
