一元取代苯甲酸酯类化合物组合合成及杀菌活性
Synthesis and fungicidal activity of mono-substituted benzoic esters
【目的】为了研究一元苯甲酸酯类化合物的杀菌活性,并能够快速筛选到杀菌活性高的新化合物,采用组合化学中的组合库合成与平行合成相结合的方法进行杀菌活性的研究。【方法】首先,将19个醇按照分子量大小分成5组,用6个取代苯甲酸其反应,构建了含有114个化合物的30个化合物库,采用菌丝生长速率法筛选得到高活性目标库。用平行合成法对3个目标库中的12个化合物进行了合成,化合物的结构均通过核磁共振氢谱(1H NMR)、电喷雾电离质谱(ESI-MS)、红外光谱(IR)确证,并测定了它们的杀菌活性。【结果】用GC-MS对10个化合物库中的化合物进行了定性和定量检测,结果表明每个化合物库中都包含有目标化合物,且化合物含量接近。杀菌活性测定结果表明,库A5B2、A6B2、A4B3、A1B4、A3B5、A4B5对10种植物病原菌的抑制率比较高,平行合成得到的化合物6g和6h对小麦根腐病原菌Bipolaris sorokiniana (Sacc.) Shoem.,水稻稻瘟病原菌Phyricularia oryzae Cav.,水稻恶苗病原菌Fusarium moniliforme Sheld.具有非常高的杀菌活性。【结论】一元苯甲酸酯类化合物具有良好的杀菌活性,采用组合化学的方法明显降低了合成与杀菌活性测定的工作量,并筛选得到了高活性化合物。
?[Goal] In order to study the fungicidal activity of mono-substituted benzoic esters, and screened out the compound of high fungicidal activity quickly, methods by using combinatorial library synthesis and parallel synthesis of combinatorial chemistry for the study of fungicidal activity.[Method] First of all, 19 alcohol were divided in to 5 groups according to the size of molecular weight, then using 6 substituted benzoic esters reacted with them, to build 30 compounds library within 114 compounds. Mycelium growth rate method was used to screen out the highly active object library. With parallel synthesis method, systhesised 12 compounds in 3 object library, and their structures were confirmed by 1H NMR, ESI-MS and IR, and tested their fungicidal activity.[Result] The result of qualitative and quantity detection of compounds in 10 compounds library with GC-MS showed that every compound library contains target compound, and their content were similar. The test of fungicidal activity showed that the libray of A5B2、A6B2、A4B3、A1B4、A3B5、A4B5 had high inhibition rate against 10 plant pathogen. The compound 6g and 6h from parallel synthesis had very high fungicidal activity against Bipolaris sorokiniana (Sacc.) Shoem., Phyricularia oryzae Cav. and Fusarium moniliforme Sheld.[conclusion] The mono-substituted benzoic esters have high gungicidal activity, the workload of synthesis and fungicidal activity testing was significantly reduced by adopting the method of combinatorial chemistry, and screened out highly reactive compound.
