Immobilization of D-Mannose Isomerase on Ion-exchange Resins and its Enzymatic Properties

从10种大孔吸附树脂及离子交换树脂中筛选出固定化效果最好的强碱性苯乙烯系阴离子交换树脂FPA90 Cl。以戊二醛为交联剂,通过先吸附后交联的方法研究D-甘露糖异构酶的固定化条件及酶学性质。结果表明,最佳固定化条件为:加酶量26 U/g树脂,吸附pH 8.5,吸附温度为28℃,吸附4 h后,置于4℃冷却30 min,加入戊二醛溶液至体系内戊二醛体积分数为0.04%,在4℃下交联2 h,最优条件下的固定化酶活回收率可达70%以上。固定化酶的最适温度为45℃,最适pH 8.0,温度稳定性和pH稳定性都优于游离酶,且固定化酶的米氏常数比游离酶高。固定化酶在重复使用8次后仍保留80.8%的酶活,表明该固定化酶具有较好的操作稳定性。

In this paper, D-mannose isomerase was immobilized on 10 types of macroporous adsorption resins and ion-exchange resins, in which FPA90 Cl performed the best immobilization effect. Using glutaraldehyde as a cross-linkage agent, the optimum conditions for the immobilization were as follows: enzyme load of 26 U/g resin, 4-hour absorption at 28℃, pH 8.5, adding the glutaraldehyde of which concentration was 0.04% and 2-hour cross-linking at 4℃. With these conditions, the recovery yield of the immobilized enzyme reached over 74%. The optimum temperature was 45℃, the optimum pH was 8.0, the temperature stability and pH stability were both better than the free enzyme. And it presented higher Michaelis constand than thefree enzyme and retained 80.8% of its initial enzyme activity after reuse for eight times, indicating that the immobilized enzyme has good operational stability.
