中枢给予卡托普利对心衰大鼠外周炎性反应的影响
Effects of Central Treatment with Captopril on Peripheral Inflammation in Heart Failure.
目的:观察心衰时下丘脑室旁核中肾素-血管紧张素系统对外周炎性反应的影响。方法:选取雄性SD大鼠28只,采用冠状动脉结扎术建立大鼠心力衰竭模型或假手术模型,通过大鼠颈部植入微型渗透压泵,侧脑室插管持续缓慢给予ACE抑制剂卡托普利(CAP,2.1μg/h,ICV)或空白对照药。4周后通过酶联免疫吸附试验(ELISA)测定血浆中IL-1β和IL-6的含量。结果:与假手术组相比,心力衰竭大鼠血浆IL-1β,IL-6水平明显升高(P<0.05)。与接受Vehicle治疗的心力衰竭大鼠相比,接受卡托普利治疗的心力衰竭大鼠血浆IL-1β、IL-6水平下降(P<0.05)。结论:心力衰竭时下丘脑室旁核中肾素-血管紧张素系统促进外周炎性反应,进一步加重心衰。
Objective: To explore the effects of angiotensin converting enzyme(ACE) inhibition in hypothalamic paraventricular nucleus (PVN) on peripheral inflammation in heart failure (HF). Methods: HF in rats was induced by left anterior descending coronary artery ligation. Sham-operated control (SHAM) or HF rats were treated with either ACE blocker Captopril(CAP, 2.1μg/h, ICV) or vehicle through intracerebroventricular (ICV) infusion for 4 weeks. Results: Compared with SHAM rats, plasma levels of IL-1β and IL-6 were increased in HF rats(P<0.05). ICV infusion of Captopril attenuated the increases in plasma of IL-1β and IL-6 induced by HF (P<0.05). Conclusion: The renin-angiotensin system (RAS) in PVN modulates peripheral inflammation which could account for pathogenesis of heart failure.
