棉纤维特异启动子LTP12驱动的基因phaB、phaC双价载体构建及其原核表达研究

聚羟基丁酸酯(PHB)具有抗皱、保暖等特性,若棉纤维中存在PHB则棉纤维保暖性和抗皱性将会提高,因此通过基因工程技术手端将聚羟基丁酸酯的合成酶基因导入棉花中,将会使棉纤维发育过程中合成PHB,提高棉纤维的抗皱性、保暖性和防水等特性,从而达到改良棉纤维的品质。本研究分离了棉纤维特异启动子LTP12和聚羟基丁酸酯合成酶基因phaB和phaC,构建了由LTP12驱动的基因phaB和phaC的双价植物表达载体pBLTPPBPC。将基因phaB和phaC在大肠杆菌BL21中进行诱导表达,并用SDS-PAGE电泳检测其表达情况。结果显示,基因phaB电泳凝胶图谱出现一条分子量约35KD的特异性普带,其大小与基因phaB的表达产物分子大小基本相符。而基因phaC经诱导后未出现特异性普带,这可能与基因phaC本身有关。
he poly-3-hydroxybutyrate(PHB) have anti-crease and warmth retention , However, the anti-crease and warmth retention of the cottonfiber would improved if it has PHB in the cotton fiber , So the PHB synthase genes was introduced cotton by Genetic Engineering Technology that the PHB was synthased in the cotton fiber, the waterproofness、 anti-crease and warmth retention of the cotton fiber was improved. In this work, promoter LTP12 for cotton fiber and genes phaB and phaC are cloned. then their plant expression Vector pBLTPPBPC drived by Specific Promoter LTP12 was constructed and the two genes were induced prokaryotic expression in Escherichia coli BL21. The expression products of the genes phaB and phaC was detected by polyacrylamide gel electrophoresis. The result showed that gel electrophoresis of gene phaB has a line of molecular weight about 35KD, the molecular weight of the gene phaB is the same size of 35KD, however, there is no special line on the gel electrophoresis of gene phaC, which may be connected with gene phaC.
