Legumain在肿瘤转移中的作用及以其为靶点的相关治疗方法的研究

Legumain是一种天冬氨酸内肽酶,属于应激蛋白。基因表达谱及组织芯片的分析研究结果表明legumain在肿瘤组织及肿瘤相关巨噬细胞(TAM)中高表达, 根据此现象而设计的legumain微基因肿瘤疫苗能够有效地靶向肿瘤细胞及TAM,从而达到抑制肿瘤的生长、新生血管生成及远端转移的作用。同时以legumain作用底物和阿霉素为主要构成部分的化疗药物legubicin能够提高药物的靶向性及药效。本文将对legumain在肿瘤发展中的作用及相关药物的开发研究进行回顾与总结。
Legumain, an asparaginyl endopeptidase, is a kind of stress proteins. The analysis of gene expression profiling and tissue array indicates that legumain is highly expressed in tumor and tumor associated macrophage (TAM). Based on this character, legumain mini-gene vaccine is developed and has been demonstrated to inhibit the growth, metastasis and angiogenesis of tumor efficiently. Also, the chemical of legubicin, which uses doxorubicin and the substrate of legumain as key components can specifically target on tumors and increase the therapeutic effect. This review focuses on legumain to introduce its function in the development of tumor and the relative progression in the therapeutic strategy of tumors.
