目的:探讨糖皮质激素对新生小鼠始基卵泡池形成及卵泡发育的影响。方法:实验分为2个组。A组(注射组):将正常出生后1 d 的雌幼C57/BL小鼠皮下注射糖皮质激素 (35 mg/kg),分别用多聚甲醛固定出生后7 d龄雌仔鼠卵巢。B组(正常对照组):正常新生小鼠,于生后第1天注射同剂量的生理盐水(35mg/kg),分别固定出生后7 d龄雌仔鼠卵巢。同时所有卵巢经HE染色,观察小鼠生殖细胞巢破裂情况和不同发育阶段的卵泡数目; 结果:注射组(A组)较对照组(B组)小鼠生长较迟滞;A组(注射组)与B组相比,7d龄小鼠始基卵泡比例较低,然而7d龄初级与次级卵泡比例无显著差异; 结论:大剂量糖皮质激素能加速新生小鼠卵母细胞巢破裂,增加卵母细胞凋亡,缩小始基卵泡池规模,可能有害于卵巢的生殖寿命。

Objective: To investigate glucocorticoid on ovarian follicular development and oocyte apeptosis in neonatal mice. Methods: A totaI of 30 SD C57/BL mice were caged together with female and male ratio of 2:1, and the neonataI female rates were divided into two groups.Group A(The neonatal mice were administered a subcutaneous injection of 35 mg/kg of DEX on D1); Group B(The control mice received injections of 35 mg/kg of NS on D1). Left ovaries from 7-day-old female pups were stereologically analyzed. The number of primordial and primary follicles and the number of secondary follicles was determined by exact counts of every fourth section encompassing whole cross-sections of the ovary. Results: In D7 ovaries,compared with group B,the ratio of primordial follicles from group A decreased remarkably(P<0.05); But as to primary and secondary follicles, There were no significant differences between group A and group B(P>0.05).Conclusion:Glucocorticoid may promote oocyte nest breakdown in neonatal mice,speed up primordial follicle assembly and development,as well as promote oocyte apoptosis,thus may be harmful to the reproductive lifespan of ovary.
