目的:探讨中国汉族冠心病患者染色体9p21区域rs2383207位点遗传变异与相邻基因CDKN2A和CDKN2B表达水平的关系。方法:选择冠心病患者91例为研究对象。采用Sequenom MassARRAY飞行时间质谱法对rs2383207位点进行基因分型,采用实时荧光定量PCR检测CDKN2A和CDKN2B基因的表达水平,采用SPSS16.0进行统计分析。结果:染色体9p21区域rs2383207位点不同基因型冠心病患者外周血中CDKN2A表达量差异显著(P<0.05),而CDKN2B基因的表达量无显著差异(P>0.05)。结论:rs2383207位点遗传变异可能是通过影响CDKN2A基因的表达影响冠心病的发生。

Objective: To explore the relationship between rs2383207 genetic variation in 9p21 region and expression levels of the adjacent genes in patients with coronary heart disease (CHD) in Chinese Han Population. Methods: We recruited 91 patients with CHD as subjects . Rs2383207 loci were genotyped with Sequenom MassARRAY time of flight mass spectrometer (TOF); gene expression levels of CDKN2A and CDKN2B were detected with Real-time Quantitative PCR. SPSS16.0 was used for result analysis. Results: Gene expression level of CDKN2A was different in peripheral blood of CHD patients with different genotype (P<0.05), while CDKN2B was not (P>0.05). Conclusions: Genetic variation of rs2383207 locus influencing the accurrence of CHD may be by influencing the expression level of CDKN2A gene.
