姜黄素对结肠癌lovo细胞增殖和黏附能力的影响
Effect of curcumin on Growth and Adhesion of Human Colon Carcinoma Cell Line Lovo
目的 观察姜黄素对人结肠癌lovo细胞的生长抑制作用。方法 以不同浓度(25 -100μmol/L) 的姜黄素作用于体外培养的lovo细胞48h,利用光镜观察各实验组癌细胞的密度与形态改变;应用MTT 方法检测细胞的生长抑制率, 细胞黏附实验检测细胞的黏附性.。结果 经过姜黄素48h 的处理后,lovo细胞出现了明显的生长抑制现象,随着姜黄素剂量的增加,贴壁细胞数量减少,死亡脱落细胞增多。初25μmol /L组外,50μmol/L 和100μmol/L 姜黄素组与阴性对照组比较均有显著性差异(P<0.05)。90, 120分钟时,细胞黏附数显示50μmol /L 和100μmol /L 姜黄素组比对照组细胞黏附数显著降低( P < 0.05 )。结论 姜黄素能抑制lovo细胞体外增殖和黏附,并呈现剂量依赖关系。
Objective:To observe the effect of curcumin on the growth and proliferation of human colon cancer cell line lovo invitro.Methods:The lovo cells were treated with 25-100μmol/L curcumin for 48 hours respectively.The morphological changes of lovo cells were observed with inverted microscope,The effects of curcumin on the growth of lovo cells were detected by MTT assay, and adhes ion ofcell line was evaluated by cell adhesion assay.Results:MTT method revealed that curcumin could inhibit the growth of lovo cells in a dose-dependent man-ner.Except low dose(25μmol/L) group,other dose curcumin treated group (50μmol/L and 100μmol /L) had significant difference compared with negative control group (P<0.05). And the growth inhibit rates of these curcumin groups had significant difference comparedwith the control group (P<0.05); MTT method aslo showed that the adhesion ability of curcumin group(50μmol /L and 100μmol/L) dramatically lower than those in the control groups at 90min and 120min respectively (P<0.05).Conclusion:Curcumin could inhibit the growth and adhesion of lovo cells in a dose-dependentmanner in vitro.
