饲料添加剂用黄芩甙提取工艺条件的优化
Optimization of Baicalin Extracting Technique in Scutellaria baicalensis Georgi
为了优化饲料添加剂用黄芩甙提取工艺,采用L9(34)正交实验设计方法,对浸提和酸沉工艺进行了优化实验研究,采用紫外分光光度法对提取物黄芩甙进行含量测定。结果表明:浸提较优工艺为A3 B3 C2,即浸提时间1.5h,浸提温度100℃,浸提pH7.0;酸沉较优工艺为A′2 B′3 C′1 D′3,即保温时间1.0h,保温温度80℃,保温pH1.0,静置时间12h。经两次正交实验设计所得优化提取工艺可行,提取率为91.56%,含量为94.12%。
In order to optimize the extracting technique of baicalin in Scutellaria baicalensis Georgi, the orthogonal experimental design was adopted in the diyestion and the acid-deposition processes, UV spectroscopic method was employed to determine the content of baicalin . Results proved that the best diyestion technique was A3B3C2, i.e. diyestion time was 1.5 h, temperature was 100℃, pH was 7.0; the best acid deposition technique was A′2B′3C′1D′3, i.e. heat preservation time was 1.0 h, temperature was 80℃, pH was 1.0, whisht placement time was 12 h. The optimized techniques mentioned above were feasible. The extraction rate was 91.56%, baicalin content was 94.12% based on the techniques.
