美国王鸽脑组织中α-突触核蛋白基因编码区的克隆与序列分析

为克隆与分析美国王鸽脑组织中α-突触核蛋白基因序列,用PCR方法从鸽脑组织cDNA中扩增出α-突触核蛋白基因编码区序列,进行质粒PCR鉴定及DNA测序。获得一条609bp的特异性片段,测序结果与原鸡参考序列同源性为93.52%,成功克隆出鸽α-突触核蛋白基因编码区序列。人、小鼠、大鼠等α-突触核蛋白基因编码区序列同源性在56.10%-96.45%内,说明存在一定的种属差异。
o clone and analyze α-synuclein gene sequence of the King pigeon brain, α-synuclein genes coding sequences were amplified by PCR, identified by the plasmid PCR and sequenced. Sequencing of the positive clones showed that the gene fragment was 609bp. Sequence analysis showed that the identity between King pigeon and Gallus gallus was 93.52%, which illustrated that α-synuclein gene coding sequences of King pigeon were cloned successfully. While the identity among Homo sapiens, Mus musculus,Rattus norvegicus and other species was 56.10%-96.45%, indicating that there is speciesdifference among them.
