甜瓜CmERFIV-3基因cDNA的克隆及功能初步分析

为了研究甜瓜ERF(ethylene-responsive factor)亚家族基因在果实成熟过程中的功能,本研究以甜瓜(Cucumis melo L.)品种河套蜜瓜为材料,克隆得到甜瓜ERF亚家组成员之一CmERFIV-3(MELO3C021306)基因cDNA,其开放阅读框(ORF)长为702 bp,编码233个氨基酸。分别构建了CmERFIV-3基因的超表达载体pPZPIV-3和RNAi载体pARTIV-3,采用农杆菌介导的瞬时表达技术将含有重组质粒的农杆菌菌液注射到成熟期甜瓜果实中。结果显示,注射超表达载体的部位与对照相比,23.3%样品出现提前变黄的表型,而注射RNAi载体的部位与对照相比,20%样品出现持绿的表型。实时荧光定量PCR结果表明,注射超表达载体部位的目的基因的表达量是对照的2.37倍,而注射RNAi载体部位的目的基因的表达量是对照的0.24倍。这些结果初步表明CmERFIV-3基因参与甜瓜果实成熟过程。
he CmERFIV-3(MELO3C021306) gene, one of the ERF subfamily members, was cloned by RT-PCR method using cDNA as template from the fruit of Cucumis melon to analyze the function of ERF subfamily genes during melon fruit ripening. The ORF of this CmERFIV-3 gene was 702 bp, which encoded 233 amino acids. The full-length of the CmERFIV-3 gene was combined with overexpression vector pPZP221 and two interfering sequences against CmERFIV-3 gene were constructed into RNAi vector pART27 respectively. These vectors were then transformed into Agrobacterium AGL1 and injected into mature green fruits of melon separately using the method of transient expression inducted with Agrobacterium. The phenotypes of melon pericarp showed that the tissue transformed by overexpression vecors turned yellow ahead of the control group which took the proportion of 23.3% and the tissue transformed by RNAi vectors maintained in green in comparison to the control group which took the proportion of 20%. The expression level of the CmERFIV-3 gene was analyzed by RT-qPCR. The results revealed the mRNA level of the CmERFIV-3 gene in the tissue transformed by overexpression vector was 2.37 times in comparison to the control group. In addition, the expression level of the CmERFIV-3 gene in the tissue transformed by RNAi vectors was reduced about to 0.24 times compared with the control group. These results indicated that CmERFIV-3 gene may have a function of acceleration during fruit ripening in melon.
