三种鳗鲡的种质鉴定及遗传多样性分析
Identification of 3 Populations of eel Genetic Diversity Analysis by Means of Random DNA( RAPD)
运用随机多态性扩增DNA(random amplified polymorphic DNA,RAPD)技术对澳洲鳗鲡(Anguilla australis)、欧洲鳗鲡(Anguilla anguilla)和日本鳗鲡(Anguilla japonica)的遗传多样性进行研究。从40个随机引物中筛选出12个引物重复性好的RAPD 随机引物,分别用于3 种鳗鲡基因组DNA 的分析,均得到1-10条大小不等的DNA条带,多数条带的分子量在400~1500 bp之间,其中S281、S286、S326、S335引物在鳗鲡群体产生的分子标记具有明显的种间特异性,单独或组合用于鳗鲡品种鉴定效果最佳。3种鳗鲡的种间遗传距离统计结果表明:澳洲鳗鲡与日本鳗鲡的遗传距离最小,为0.125,欧洲鳗鲡与澳洲鳗鲡的遗传距离最大,为0.500,欧洲鳗鲡与日本鳗鲡的遗传距离0.250。本研究结果为鳗鲡品种的快速鉴定和种质资源的研究奠定基础。
RAPD (random amplified polymorphic DNA) markers were applied to detect the genetic diversity and molecular evolution of 3 ell populations included Anguilla japonica, A. anguilla and A. australis. 12 arbitrary primers screened out of 40 random primers were adopted to analyze the polymorphism in RAPD profiles, the random primers S281, S286, S326 and S335 produced different bands in the 3 eel populations, which could be used to identify the stock. The 12 primers could generate 1-10 amplification bands for each species with fragments sized from 300 to 2200 bp. The genetic distances were 0.125 between A. australis and A. japonicus, 0.500 between A. anguilla and A. australis, 0.250 between A. anguilla and A. japonicus. The results provides a basis for the rapid identification of Anguilla Shaw varieties and germplasm resources study.
