分别采用酶消化法分离获取兔成骨细胞和骨髓诱导法分离获取兔破骨细胞,对在体外培养的成骨细胞(碱性磷酸酶染色和钙化能力分析)和破骨细胞(抗酒石酸酸性磷酸酶染色、骨吸收功能、肌动)进行鉴定与功能观察。结果显示:所培养的成骨细胞碱性磷酸酶染色阳性,具有较好的钙化能力; 破骨细胞表现为多核巨细胞外观,抗酒石酸酸性磷酸酶染色阳性, 具有良好的骨吸收功能,胞周形成肌动蛋白环。提示成功分离获得成骨细胞和破骨细胞,为进一步的成骨细胞-破骨细胞间信号通讯研究打下基础

In this study, rabbit’s osteoblasts and osteoclasts were harvested through the enzyme digestion method and bone-marrow-cells induction approach, respectively. Then cultured osteoblasts were assayed by alkaline phosphate (ALP) staining , mineralized function and osteoclasted were identified through tartrate-resistant acid phosphatase (TRAP) staining , bone resorption assay and F-actin ring observation. The results showed that osteoblasts were available with ALP positive and good mineralized ability and osteoclasts were obtainable with TRAP positive, fine bone resorption ability , obvious F-actin ring around cytoplasm. It is suggest that the target osteoblast and osteocalst for following experiments are available.
