目的:探讨细胞因子信号传导抑制蛋白1(Suppressors of cytokine signaling, SOCS-1)在糖尿病大鼠肾组织炎性损伤中的作用。方法:雄性CD-1小鼠随机分为四组,对照组、糖尿病组、SOCS-1质粒转染组和空载体转染组。腹腔注射链脲佐菌素(Streptozotocin, STZ)诱发糖尿病小鼠模型,转染组在成模后给予尾静脉注射SOCS-1质粒或空质粒载体,每隔7天注射一次。4周后收集标本,检测各组动物的血糖、24h尿蛋白;Western 蛋白印迹法检测肾组织中SOCS-1蛋白的水平。流式细胞术检测肾组织中巨噬细胞标志蛋白CD68的水平; ELISA检测肾组织中白细胞介素-1β(Interleukin-1β, IL-1β)和转化生长因子-β1(Transforming growth factor-β1, TGF-β1)的水平。结果:过表达SOCS-1能抑制糖尿病大鼠肾组织中CD68、IL-1β和TGF-β1的表达、降低糖尿病大鼠尿蛋白排泄,但对血糖无影响。结论:过表达SOCS-1能通过减轻糖尿病大鼠肾组织炎症反应从而缓解肾损伤。

Objective To investigate the role of SOCS-1 on diabetic renal injury. Methods Male CD-1 mice were randomly divided into four groups: control group, diabetic group, empty plasmid vector transfection group. SOCS-1 plasmid transfection group. Mice received an intraperitoneal injection of STZ at a dose of 150mgokg-1 body weight. Transfection mice received an injection of SOCS-1 plasmid or empty vector and every 7 days thereafter. At 4 weeks after STZ injection, specimens were collected to detect the blood glucose and 24 hours urine protein. Western blotting analysis were used to determine the expression of SOCS-1. The expression of CD68 was detected by flow cytometry. The secretion levels of IL-1β and TGF-β1 were detected by ELISA. Results Overexpression of SOCS-1 in kidney ameliorated excretion of urine protein and the expression of CD68, IL-1β and TGF-β1. Conclusion Overexpression of SOCS-1 can reduce the inflammatory response in renal tissue of diabetic rats and relieve renal injury.
