辅助调节因子是类固醇激素和配体结合后募集的转录调节因子,对类固醇激素受体的转录活性具有上调或下调作用,从而调节细胞的增殖、分化、凋亡等生物学表型。在本文研究的类固醇受体辅助调节因子中,ARA55的表达水平在前列腺癌细胞系中明显低于非癌细胞系;在前列腺癌组织中表达水平低于癌旁组织。提示,ARA55的表达下调与前列腺癌相关。MTT结果表明,ARA55过表达可以抑制DU-145细胞的增殖。实时PCR和ELISA的结果显示,ARA55过表达可以上调TGFβ1的表达;而且ARA55对DU-145增殖的抑制作用可以部分被TGFβ1中和抗体阻断,提示,ARA55抑制增殖的作用是部分通过TGFβ1而实现,但可能还有其他途径。ARA55在前列腺癌中低表达;ARA55可上调TGFβ1的表达,ARA55又可通过促进TGFβ1的表达部分抑制前列腺癌细胞系DU-145的增殖。提示ARA55和TGFβ1在前列腺癌中进程中发挥作用。

Steroid hormone receptor (SHR) co-regulators (CoR) are the transcription factors recruited onto the promoter when steroid hormone binds to receptors, CoR may regulator target gene expression and activity in cells so that CoR may regulator cell proliferation, differentiation and apoptosis. In this paper we find that ARA55 expression decrease in prostate cancer cells and tissues. Overexpression of ARA55 can decrease proliferation of DU-145 cells. Real time RT-PCR and ELISA results show that overexpression of ARA55 can upregulate the expression of TGFβ1.TGFβ1 neutralizing antibody can partially block ARA55 desregulation of DU-145 proliferation. These results indicate ARA55 down regulates in DU-145 prostate cancer cell proliferation partially through TGFβ1. ARA55 expression decreased in prostate cancer. Overexpression of ARA55 inhibits DU-145 proliferation partially by upregulation of TGFβ1, while other pathways may be involved. These results uncover novel roles and mechanisms of ARA55 and TGFβ1 in the prostate cancer.
