水稻传统品种月亮谷抗感品系与稻瘟病菌的群体互作
Population interaction between Magnaporthe oryzae and resistant or susceptible pure lines derived from rice landrace Acuce
以合系22-2为对照,利用15对稻瘟菌SSR引物对田间自然发病的月亮谷自然群体、四个月亮谷单粒传不同抗感表型的纯系及合系22-2上分离到的540个稻瘟菌菌株进行遗传多样性分析。结果显示,稻瘟病菌群体遗传分化明显(Ht=0.15±0.13,I=0.27±0.19, Fst=0.30),来自不同品种(品系)的稻瘟病菌群体遗传差异较大(p<0.05);聚类分析结果表明,在遗传相似系数0.9水平下,分离自合系22-2、月亮谷抗病纯系上的稻瘟菌只有1个宗谱,而来自感病月亮谷纯系和月亮谷自然群体的稻瘟病菌存在4-8个宗谱,说明抗病品种(品系)对稻瘟病菌产生了选择压力;对单病斑上分离的稻瘟病菌聚类分析结果显示,来自不同品种(品系)的病菌存在1-5个不同的宗谱,说明在同一定殖系统中存在不同遗传来源的菌株;540个稻瘟病菌的AMOVA分析结果表明,78%的变异来源于群体内。
Population genetic characteristic of 540 M. oryzae isolates, pathogenic to rice Hexi 22-2, landrace Acuce pure lines with susceptible or resistant phenotype and natural population, were analyzed by using 15 SSR markers. The results revealed that all the isolates distinctly diversified into individual lineage (Ht=0.15±0.13,I=0.27±0.19, Fst=0.30), significant variation of inter- varietal and lines strains was observed(p<0.05). With cluster analysis, at similarity coefficient 0.9 level, there are only 1 dominant lineages in the isolates population from resistant Hexi 22-2 and resistant Acuce pure lines, however, 4-8 lineages was observed on blast-susceptible lines. This result indicated that resistant modern variety and blast-resistant Acuce pure lines exert selection pressure on M. oryzae population evolution. Further, for clarifying isolates diversification on single lesions, the isolates from single lesion of individual material was analyzed through clustering method, ten isolates from each panicle was clustered into 1-5 lineages, it impled that one colonization system could consist of isolates with sophisticated genetic source. AMOVA result based on 540 isolates analysis showed that intra-population variance contributed 78% variance to the population genetic variance of M. oryzae
