玻璃化法处理的同种异体动脉移植的实验研究

目的 通过比较玻璃化法和传统冷冻法保存处理的同种异体血管移植后效果,从而找出一种更为实用且制作简便的血管处理方法。方法 健康青紫兰兔96只,手术切除双侧股动脉, 根据不同体外预处理血管方法将实验分为3组:A新鲜血管自体移植;B冷冻+辐照预处理血管同种异体移植;C玻璃化+辐照预处理血管同种异体移植。血管移植后1周、4周、8周、12周,每组取试验动物6只进行腹主动脉数字减影血管造影、扫描电子显微镜、组织病理等方法,观察移植血管通常率、动脉瘤形成情况及组织形态学变化。结果 移植后12周,A组累计通畅率为91.67%,B组为70.83%,C组为79.2%,A组与C组、B组与C组比较,P〉0.05,差别无统计学意义。A组与B组比较P<0.05差别有统计学意义。组织病理学检查显示,C组处理的同种异体血管内膜及中层平滑肌增生较B组轻,管腔狭窄不明显,炎症反应较轻。 结论 经玻璃化法+辐照保存的同种异体血管,制作程序简单,移植血管通畅率高,组织反应轻,是一种比较理想的同种异体血管体外处理方法。
Objective To compare vitrification and freezing to preserve the traditional treatment of allograft vascular effects after transplantation, and to find a more practical and easy production of vascular treatment methods.Methods Health bruising blue rabbit 96, surgical resection of bilateral femoral artery, according to different methods of in vitro vascular preconditioning experiment were divided into 3 groups: A new vascular autograft; B irradiation pretreatment frozen allograft vascular; C glass+irradiation pretreatment of vascular allografts. Vascular grafts after 1 week, 4 weeks, 8 weeks, 12 weeks, each taking six experimental animals for abdominal aortic digital subtraction angiography, scanning electron microscopy, histopathology and other methods to observe the vascular graft is usually the rate of aneurysm formation Morphological changes and organization.Results The results of 12 weeks after transplantation, A group the cumulative patency rate of 91.67%, B group was 70.83%, C group was 79.2%, A group and C group, B group and C group, P> 0.05, difference not statistically significant. A group and B group, P <0.05 difference has statistical significance. Histopathological examination revealed, C group treatment of allograft vascular smooth muscle proliferative endometrium and the middle group than the B light, the lumen stenosis was not obvious, less inflammatory response. Conclusion The vitrification irradiation preservation of allograft blood vessels, making the procedure easy, high patency rate of grafts, tissue reaction to light, are an ideal way to deal with allogenic blood vessels in vitro.
