N端结构域在Galectin-3诱导T细胞凋亡中的作用

半乳糖凝集素-3(Galectin-3,Gal-3)是一种能结合β-半乳糖苷的凝集素,由N端的前导结构域(LD)和胶原样重复结构域(RD)以及C端的碳水化合物识别结构域(CRD)构成。Gal-3能诱导T细胞凋亡,与肿瘤免疫逃逸有关。目前尚不明确各种结构域与其凋亡活性的关系。本文利用FITC-AnnexinV/PI双染、western blot(检测caspase-3活化)和流式细胞术(检测Sub G1细胞)等方法,通过比较全长和一系列截短蛋白(G13-250、G69-250、G111-250和G1-108)诱导T细胞凋亡的能力,研究了各种结构域的作用。结果表明,CRD是诱导凋亡必需的结构域,但是单独CRD不能诱导凋亡,必须含有一部分N端结构。N端的RD对功能的发挥至关重要,而LD则与诱导凋亡无关。利用N端和C端特异性抑制剂进一步证实了上述结论。本文首次报道了N端结构域在诱导T细胞凋亡中的作用,进一步明确了Gal-3的作用机制。研究结果对Gal-3抑制剂的开发具有重要的指导意义。
Galectin-3 (Gal-3), a β-galactoside-binding protein, consists of an N-terminal leading domain (LD) followed by a collagen-like repeating domain (RD) that is connected to a carbohydrate recognition domain (CRD) at the C-terminus. Gal-3 can induce T-cell apoptosis and is associated with tumor escape from immune surveillance. The relationship between Gal-3 structure and its apoptotic activity remains elucidative. Here we investigated the functions of each structural domain by comparative analysis of the full length protein with various truncated proteins (G13-250、G69-250、G111-250 and G1-108) using the approaches of FITC-AnnexinV/PI double staining, western blot (for activated caspase-3) and flow cytometry (for the Sub G1 cell population). The results showed that the CRD is prerequisite for Gal-3's pro-apoptotic action. However, the CRD alone is unable to induce apoptosis and it must connect to a segment of the N-terminal tail for function. While the LD is not required, the RD plays a pivotal role in initiating apoptosis. Further studies with inhibitors specific to the N-terminal tail and CRD confirmed these results. This study reveals the functions of the LD and RD in Gal-3 induced T-cell apoptosis and is useful for the development of galectin-3-targeting anti-cancer therapeutics.
