越来越多的证据表明,E3泛素连接酶在癌症发展中发挥着重要作用。Nedd4类E3泛素连接酶家族有九个成员,均具有一个相似的结构,包括在N端的C2结构域,在蛋白结构中间2-4个WW结构域,和位于C端的HECT结构域。Nedd4家族在癌症中发挥作用得到越来越多的数据支持。在食管鳞状细胞癌中,Smurf2 过表达;在前列腺癌和乳腺癌中WWP1过表达;前列腺癌及膀胱癌中Nedd4过表达;胰腺癌中Smurf1过表达。Nedd4类E3连接酶调节泛素-蛋白酶体复合物的组装、溶酶体或蛋白酶体降解、多种蛋白质核易位等,因此能够通过调节重要的信号通路,如TGFβ、EGF、IGF、VEGF、SDF-1、TNFα等,参与肿瘤发生发展。此外,部分Nedd4类E3连接酶成员能够直接调节各种癌症相关的转录因子,比如Smad、p53、KLF、RUNX、Jun家族等。同时多个Nedd4类E3连接酶具有独立活性。此外,Nedd4类E3连接酶在肿瘤细胞中其本身能通过磷酸化、泛素化、转位及转录调控而调节。因其调节底物及被调节的上游蛋白非常复杂,所以探讨Nedd4类E3连接酶在肿瘤中的作用,仍需要更多的研究及数据支持。了解Nedd4类E3连接酶在肿瘤中的作用,能够促进鉴定及发现新的生物标记,并促进对肿瘤药物的研发。

More and more evidences indicated that E3 ubiquitin ligases played an important role in cancer development. So far as we know, there are nine members in Nedd4-like E3 family, with a similar structure including a C2 domain at the N-terminus, two to four WW domains in the middle, and a HECT domain at the C-terminus. It was reported that Nedd4 family were involved in tumorgenesis and development, such as WWP1 in breast and prostate cancer, Nedd4 in bladder and prostate cancer, Smurf2 in esophageal squamous cell carcinoma and Smurf1 in pancreatic cancer. Besides, Nedd4 family take parts in lysosomal or proteasomal degradation, ubiquitin-mediated protein trafficking, and nuclear translocation of multiple proteins, further on, they modulate important signaling pathways involved in tumorigenesis and development, like EGF, IGF, TGFβ, VEGF, TNFα, and SDF-1. Additionally, several members of Nedd4 family regulate various cancer-related transcription factors directly, like the Smads, KLF, RUNX, p53, and Jun. Moreover, multiple E3 of Nedd4 family shows ligase independent function. Furthermore, Nedd4 family is frequently regulated by ubiquitination, phosphorylation, translocation, and transcription in cancer cells. Since the regulation and biological output of these E3 ligases is a complex process, study of the role of these E3s in cancer development need more evidences. However, understanding the potential role of these E3 may facilitate the identification and development of drug and new biomarkers in human cancer.
