中国妇女上皮性卵巢癌中AIB1 CAG/CAA重复序列多态性的研究

目的:探讨卵巢上皮性癌中AIB1基因中CAG/CAA重复序列的多态性,以了解AIB1基因中CAG/CAA重复序列的多态性能否作为预测上皮性卵巢癌发生的一项高危因子。方法:以PCR法检测24例正常卵巢、25例卵巢良性肿瘤、19例卵巢交界性肿瘤及69例上皮性卵巢癌中AIB1 CAG/CAA重复序列多态性,观察结果并对不同等位基因进行测序。结果:1)中国妇女AIB1 基因中CAG/CAA三核苷酸重复序列的重复次数为26~29,最常见的为29。纯合子29/29和杂合子28/29是最常见的基因型,分别占35.77%(49/137)和49.64(68/137)。2)AIB1 CAG/CAA重复序列≤28者分别占卵巢上皮性癌组、卵巢交界性肿瘤组、卵巢良性肿瘤组和正常卵巢组的14.49%(10/69)、10.53%(2/19)、4.0%(1/25)、0,其组间差异在统计学上均无显著性(P>0.05)。3)相关性分析显示,AIB1 CAG/CAA重复序列多态性与卵巢上皮性癌的病理类型、临床期别、病理分级和淋巴结转移无相关性(P>0.05);。结论:AIB1 CAG/CAA三核苷酸重复序列的重复次数≤28不能作为预测中国妇女卵巢上皮性肿瘤发病的高危因素。AIB1 CAG/CAA重复序列多态性与卵巢上皮性癌的病理类型、临床期别、病理分级和淋巴结转移无关,但尚需大样本研究证实。
o analysis the relationship between AIB1 CAG/CAA repeat polymorphism and epithelial ovarian cancer. 69 epithelial ovarian cancers, 19 borderline ovarian tumors, 25 benign ovarian tumors and 24 normal ovarian specimens were included in this study. PCR was used to examine AIB1 genetypes. Typical genotypes were selected and sequenced.1) The AIB1 CAG/CAA repeat length in Chinese women ranged from 26 to 29, with the most common repeat length being 29. Homozygous 29/29 and heterozygous 28/29 were the most common genotypes, with 35.77% (49/137) and 49.64% (68/137) harboring these genotypes, respectively.2) The frequency of a short AIB1 genotype (with ≤28 CAG/CAA repeats ) in epithelial ovarian cancers、borderline ovarian tumors、benign ovarian tumors and normal ovarian specimens was 14.49%(10/69)、10.53%(2/19)、4.0%(1/25)、0, respectively. There is no statistic difference in those samples(P>0.05).3) No difference of AIB1 CAG/CAA repeat polymorphism was found according to the histological differentiation, FIGO stagings, grades and lymph node involvement(P>0.05).Based on the findings in this study, it is suggested that short AIB1 genotypes (with ≤28 CAG/CAA repeats) are not a strong predictor of risk factors of epithelial ovarian cancer in Chinese women. There is no evidence that AIB1 CAG/CAA repeat polymorphism is related to the histological differentiation, clinical staging, grades and lymph node involvement. Considering our study is small, the results need to be convinced by studies in more samples.
