随着发病率的提高,结肠癌正严重威胁着我国人民的身体健康,除了传统的治疗方式以外,利用益生菌预防、缓解结肠癌的发生可能是一种有效的干预途径。【目的】结合动物模型实验结果,构建能够在体外筛选具有抑制结肠癌发生功能植物乳杆菌的体外模型,并对植物乳杆菌缓解结肠癌发生的机制进行初步探究。【方法】利用HT-29作为细胞模型评价植物乳杆菌对结肠癌细胞生长的抑制率以及对结肠癌相关信号通路的影响,并初步筛得具有抑制结肠癌发生潜力的植物乳杆菌;利用结肠癌小鼠作为动物模型,分别以鼠李糖乳杆菌GG株(L.GG)和大肠杆菌作为阳性、阴性对照,测定植物乳杆菌对结肠肿瘤数量、血清细胞因子浓度、结肠组织癌症相关信号通路和粪便中短链脂肪酸浓度的影响以验证所植物乳杆菌在体内抑制结肠癌发生的能力。【结果】CCFM420具有良好的结肠癌细胞增殖抑制能力和调控癌症相关信号通路的能力,同时也显著地减少了模型小鼠结肠肿瘤数量,减轻了体内炎症反应,并对集体癌症相关通路具有积极的调控作用。【结论】CCFM420具有潜在的抑制结肠癌发生的能力,其可能通过下调肠道内丁酸水平,从而下调Notch通路部分信号分子,进而抑制结肠癌的发生。

olon cancer is a serious threat to the health of chinese people with the increase in morbidity. In addition to the traditional treatment, the use of probiotics to prevent and alleviate the occurrence of colon cancer may be an effective intervention. [Objective] construct an in vitro model of Lactobacillus plantarum with inhibitory effect on colon cancer. base on the experimental results of animal model. Explore the mechanism of L. plantarum to alleviate the occurrence of colon cancer. [Methods] The effect of L. plantarum on the growth of colon cancer cells and the effects on the signal pathway of colon cancer were evaluated with HT-29 cell model. Strains with the potential of inhibiting colon cancer was obtained. The colorectal cancer mice were used as animal models. L. rhamnosus GG strain and Escherichia coli were used as positive and negative control. The number of colorectal cancer cells, serum cytokine concentration, colorectal cancer-related signal pathway and fecal Short chain fatty acid concentration was evaluated to confirm the ability of L. plantarum to inhibit colon cancer in vivo. [Results] L. plantarum CCFM420 has a good ability to inhibit colon cancer cell proliferation and regulate cancer-related signaling pathway. This strain significantly reduced the number of colon cancer mice, reduced the body's inflammatory response and regulated the body's cancer-related pathways. [Conclusion] CCFM420 has the potential to inhibit the occurrence of colon cancer. This strain may down regulated the Notch signaling pathway by reducing the intestinal levels of butyric acid, thereby inhibiting the occurrence of colon cancer.
