在哺乳动物中,印记基因对胚胎发育、出生后的生长和母性行为具有重要的影响。印记基因的异常表达将导致生长发育异常及包括肿瘤在内的多种疾病。因此,对基因组印记展开研究,分离和鉴定更多的印记基因,无论是对于遗传发育等科学领域基础理论的发展,还是对于医学领域有关疾病机理的研究都具有重要的意义。基因组印记研究的首要任务是鉴定新的印记基因。因此,本研究根据印记基因成簇分布的特点,将小鼠1号染色体Zdbf2-Gpr1印记区内的Adam23基因作为侯选印记基因,利用基于SNP标记的RT-PCR产物直接测序法对该基因进行印记分析。结果表明,Adam23基因优先表达父本等位基因,表现为不完全印记,具有印记区边界基因的印记特征。

Imprinted genes play important roles in fetal growth, development and maternal behavior in mammals. The correct dosage of imprinted genes is implicated in abnormalities of development and many diseases including cancer. Therefore, it is necessary to research genomic imprinting and identify novel imprinted genes for not only the developments of developmental genetics, also the study of mechanisms of related diseases. The main task of the study of genomic imprinting is the characterize of new imprinted genes. In the paper, imprinting status of the Adam23 gege in Zdbf2-Gpr1 imprinting domain on chromosome 1 were analysed as candidate imprinted genes based on clusters features of imprinted genes by direct sequencing of SNPs approach. The results of imprinting analysis showed that the gene was preferential paternally expressed in the mouse. That is, Adam23 was a gene of incomplete imprinting and characteristic of the genes located on the terminal of imprinting domain.
