UCH-L3在小鼠腹水型肝癌高、低淋巴道转移株中的表达
Expression of UCH-L3 in mouse hepatocarcinoma ascites cell lines Hca-F(highly metastatic) and Hca-P(low metastatic)
背景与目的:转移是导致大多数肿瘤患者死亡的主要原因,肿瘤转移机制及防治已成为当今重要的研究课题。Hca-F和Hca-P是一对高度同源且淋巴道转移能力显著不同的小鼠腹水型肝癌细胞,其中Hca-F为高转移细胞株,Hca-P为低转移细胞株。UCH-L3属泛素C-末端水解酶家族,泛素系统参与细胞周期、增殖及凋亡等生理病理过程。UCH-L3与肿瘤的关系研究甚少,目前尚未见有其与肿瘤转移相关联的报告。本研究观察UCH-L3基因编码蛋白在小鼠腹水型肝癌高、低淋巴道转移株(Hca-F、Hca-P)中的表达水平,分析它在这两个细胞株中表达的关系及意义,以进一步探讨UCH-L3基因在肿瘤转移过程中的作用及相关的分子病理学机制。方法:以Hca-F和Hca-P为研究对象,采用免疫细胞化学、Western Blot和流式细胞仪检测UCH-L3蛋白在Hca-F和Hca-P两细胞株的表达。结果:免疫细胞化学显示,UCH-L3蛋白在Hca-F、Hca-P细胞中均主要定位于胞浆;Western Blot及流式细胞仪结果均显示UCH-L3蛋白在Hca-F中的表达量明显低于Hca-P(P<0.01和P<0.001)。结论:UCH-L3蛋白可能与肿瘤淋巴道转移相关,为研究肿瘤的淋巴道转移机制奠定了基础。
BACKGTOUND & OBJECTIVE: Most of patients with cancer died from its metastasis, the mechanism of metastasis and prevention and therapeutics have been an important issue in tumor domain. Hca-F(highly lymphatic metastasis potential) and Hca-P(low metastasis) are a pair of syngeneic mouse hepatocellular cell lines which present different potential of lymphatic metastasis. UCH-L3 belongs to the ubiquitin C-terminal hydrolase family. There are little reports concerning the role of UCH-L3 in tumor, especially regarding its correlation with tumor metastasis. In our study, the expression of UCH-L3 were detected in Hca-F and Hca-P and analyzed the relationship between the two tumor cell lines in order to explore the role of encoding protein of UCH-L3 genes in tumor metastasis and its relevant molecular mechanisms. METHODS: the expression of UCH-L3 protein were detected in Hca-F and Hca-P tumor cells by immunocytochemistry, Western Blot and flow cytometry. RESULTS: Immunocytochemistry manifests that UCH-L3 was localized to the cytosol between the two tumor cells. Western Blot and flow cytometry indicate that the expression of UCH-L3 was lower in Hca-F than in Hca-P (P<0.01, P<0.001) .CONCLUSION: The expression of UCH-L3 may be related to the lymphatic metastasis.
